摘 要:目的:探讨蛇毒α-神经毒素对海洛因复吸大鼠脑神经元超微结构的影响。方法:建立海洛因成瘾、脱毒,复成瘾、脱毒,再成瘾、脱毒3个阶段大白鼠模型,动物设阳性对照组、美沙酮-丁丙诺啡-可乐定联合治疗组,高、低剂量蛇毒-神经毒素组共4个组。电镜下观察各组大鼠脑神经元超微结构变化。结果:海洛因复吸大鼠脑神经元出现变性、凋亡、胀亡等超微结构变化,阳性对照组随着复吸次数增加,脑神经元病变百分比依次增加;高剂量蛇毒治疗组脑神经元超微变化与美沙酮-丁丙诺啡-可乐定治疗组相近;低剂量蛇毒治疗组脑神经元超微变化与阳性对照组相近。结论:海洛因复吸大鼠脑神经元出现广泛性超微病理结构改变,随复吸次数增多而病变加重;高剂量蛇毒α-神经毒素能减轻海洛因复吸大鼠脑病理损害。Objective:To investigate the effect of a-neuronal dotoxin of snake venom on the neuronal ultrastructures in the brains of the heroin re-addicted rats. Methods:We set up heroin addiction and re-addiction models with the processes of heroin addiction-abstaining treatment-heroin re-addiction-re-abstaining treatment -heroin addiction for the third time-abstaining theatment for the third time. Rats were divided randomly into 4 groups: the control group, Methadone-Buprenorphine-clonidine group, high and low dose of α-neuronal dotoxin of snake Venom group. The changes of the neuronal ultrastructures in the brains of the rats from all groups were observed under electronic microscope. Result:Degeneration, apoptosis and oncosis were found in the neuron of the heroin re-addicted rats' brains. The changes of neuronal ultrastructures of control group increased with the increase of re-addicting times. The changes of the neuronal ultrastrnctures in the brains of the high dose of α-neuronal dotoxin of snake Venom group were similar to the those of Methadone-Buprenorphine-Clonidine group. The changes of the neuronal ultrastructures in the brains of the low dose of α-neuronal dotoxin of snalke venom were similar to those of the control gronp. Conclusion: widespread lesions have been found and were more severe in the re-addicting times. High dose of α-neuronal dotoxin of snake venom could attenuate the neurn lesions.
