摘  要:目的研究普伐他汀对激素性坏死股骨头内骨形成蛋白-2(BMP-2)表达的影响。方法54只新西兰白兔随机分为正常组、模型组和普伐他汀治疗组,每组18只。模型组和普伐他汀治疗组应用大肠杆菌内毒素及甲泼尼龙琥珀酸钠造成股骨头坏死模型,普伐他汀治疗组于造模后4周开始普伐他汀灌胃,正常组和模型组灌服等容量蒸馏水。分别于喂药后第4、8、12周分批处死动物,实时荧光定量RT-PCR检测股骨头内BMP-2mRNA的表达。结果模型组和普伐他汀治疗组各时相点BMP-2的mRNA表达水平均低于正常组,但普伐他汀治疗组喂药后第4、8、12周的表达量明显高于模型组,差异具有显著性(P<0.01)。结论普伐他汀可诱导激素性坏死股骨头内源性BMP-2mRNA的表达。Objective To investigate the effect of Pravastatin on the expression of bone morphogenetic protein-2 ( BMP-2) of steroid induced necrosis of femoral. Methods 54 New Zealand rabbits were randomly divided into normal control group,model group and treatment group ( n = 18 for each group). All rabbits in model group and treatment group were injected with lipopoly-accharide and Methylprednisolone for the establishment of femoral necrosis model. Animals in treatment group were treated with pravastatin at the 5th week, and the same volume of distilled water was given to normal control group and model group. The rabbits were sacrificed in each group at the 4th,8th and 12th week respectively. Quantitative real-time PCR was performed for detecting the expression of BMP-2 mRNA in femoral head. Results In model group and treatment group, BMP-2 mRNA expression is lower than that of normal control group at each time point, but the expression of treatment group is significantly increased compared with model group after treatment ( P 〈 0.01 ). Conclusion Pravastatin can induce the expression of endogenous BMP-2mRNA in femoral head of steroid-induced necrosis.
