摘  要:目的了解神经营养素3(NT-3)和受体TrkC在人胚脊髓发育过程中的定位分布。方法用4、5、6周龄3个时段的人胚进行免疫组织化学ABC法染色以了解不同时段NT-3和受体在人胚脊髓发育中的定位表达和分布情况。结果在人胚胎脊髓发育的3个实验段,NT-3阳性颗粒主要分布于胞质,其受体TrkC主要分布于胞膜;发育过程中NT-3及其受体在室管膜层-套层-边缘层3条带上呈现不同程度的免疫阳性反应。结论NT-3及其受体TrkC在不同时期的人胚胎脊髓发育中进行表达,提示NT-3可能诱导神经干细胞分裂增殖,并协同其它生长因子促进轴突的生长,保证神经元胞体的存活。Objective To investigate the expression and the distribution of the Neurotrophin-3 (NT-3) and its receptor TrkC in the development of spinal cord of human embryo. Methods The human embryos in three phases(4W, 5W, 6W)and immunocytochemistry ABC staining with anti- NT-3 and anti-TrkC antibody were used to observe the distribution of NT-3 and TrkC in the spinal cord of human embryo. Result In the three phases of human embryonic development, NT-3 positive particle mainly distribute in cytoblastema, TrkC mainly distribute in the cell membrane. During the embryonic development, NT-3 and TrkC showed a positive immune reaction to a greater or less extent in ependymal zone mantle zone marginal zone. Conclusion NT-3 and its receptor TrkC expressed in different phases of human embryonic development. The results showed that NT-3 could promote the proliferation of neural stem and maintain their survival.
