摘 要:目的探讨PARP-1在适应性反应中的作用。方法低剂量甲醛预处理人胚肺成纤维细胞(MRC-5)后,观察细胞对高剂量甲醛攻击的适应性反应。在MTT法检测细胞存活率的基础上,荧光定量PCR技术测定不同组细胞中PARP-1及其相关因子表达的变化。结果用低剂量甲醛预刺激可提高细胞对继后大剂量甲醛攻击的抵抗力;低剂量甲醛能引起PARP-1及其相关因子表达增加,产生适应性反应时这种增加尤为突出,其中PARP-1的表达量是对照组的147倍。结论低剂量的甲醛可使PARP-1活化,进而调节其相关因子DNA-PK、P53、NF-κB的表达,促使细胞免疫功能增强,启动适应性反应的产生。Objective To investigate the role of(PARP-1 ) in adaptive response. Methods To observe the adaptive response of cells to high dose of formaldehyde after pretreatment with low dose of it. Based on cell survival rate detected by MTT method, real-time PCR was used to detect the expression change of PARP-1 and its correlation factors in various cells, Results Pretreatment with formaldehyde of 0.01 mmol/L increased the tolerance of cells to the attacking dose of 1 mmol/L of it. The expression of PARP-1 in cells treated with low dose of formaldehyde was 16,97 times of the control group, and in pretreatment group, 147 times, Conclusion Low dose of formaldehyde can activate PARP-1, which can detect the degree of DNA damage and active DNA repair enzyme DNA-PK, 1953, NF-κB et al, Thus corresponding response can be made to initiate the cell adaptive reaction,
