摘  要:目的:研究六味地黄汤及其配伍对衰老血瘀模型大鼠血栓形成的作用及机理。方法:建立衰老血瘀大鼠模型,观察六味地黄汤及其配伍对体外血栓形成的影响,测定血小板聚集、6-K-PGF1α/TXB2、ET、Fg、t-PA/PAI、自由基等以探讨其作用的可能机理。结果:六味地黄汤全方能抑制模型大鼠血小板聚集率的升高,降低Fg,减少内皮细胞分泌TXB2、PAI、ET,增加6-K-PGF1α和t-PA的水平,提高SOD活性,降低MDA和脂褐质的含量,抑制血栓形成。结论:六味地黄汤通过改善模型大鼠内皮细胞分泌功能,调节凝血和纤溶系统,改善自由基代谢,而达到抑制血栓形成的目的。Objective: To study the mechanism of anti-thrombosis in aged rats with blood stasis treated with Liuwei Dihuang Decoction(LDD) and the different combinations of its components Methods: In aged rats with blood stasis, the effect of LDD and the different combinations of its components on thrombosis was observed in vitro. And its potential mechanism was explored by measuring platelet aggregation, 6-k-PGF1α/TXB2, endothelin(ET), fibrinogen(Fg), t-PA/PAI and free radicals. Results: LDD could inhibit thrombosis by repressing platelet aggregation, lowering Fg, reducing the secretion of TXB2, PAI and ET from endothelial cell and increasing the content of 6-k-PGF1α and t-PA, enhancing superoxide dismutase (SOD) activity, decreasing the content of malondialdehyde(MDA) and lipofuscin. Conclusion: LDD can inhibit thrombosis by improving secretory function of endothelial cell, regulating blood coagulative and fibrillolytic system and meliorating the metabolism of free radicals.
