摘  要:目的建立测定新雪颗粒中穿心莲内酯和脱水穿心莲内酯的含量的方法。方法采用HPLC法,色谱柱为Diamonsil—C18柱,甲醇-水(体积比55:45)为流动相,流速1.0mL/min,检测波长225nm和254nm,柱温为30℃。结果穿心莲内酯在0.06387~1.597μg范围内、脱水穿心莲内酯在0.06534~1.634μg范围内呈良好的线性关系,平均回收率分别为100.2%和98.0%。结论所用方法简便、准确,能有效的控制该制剂的质量。Objective To establish a method for the content determination of andrographolide and 14-dexo-11, 12- didehydroandrographolide in Xinxie granula. Methods The analysis was performed on an HPLC Diamonsil C18 column (200 mm × 4.6 mm,5μm). The mobile phase consisted of methanol-water (55:45). The flow rate was 1. 0 mL/min. The optimum detection wavelengths were 225 nm for andrographolide and 254 nm for 14-dexo-11,12-didehydroandrographolide. Results The calibration curves showed good linear response ranged from 0. 06387 to 1. 597μg for andrographolide and 0. 06534 to 1. 634μg for 14-dexo-11,12- didehydroandrographolide, The average recoveries for andrographolide and 14-dexo-11, 12-didehydroandrographolid were 100. 2% (RSD = 0. 48%, n = 6) and 98.0% ( RSD = 0. 73 %, n = 6) , respectively. Conclusion This method is simple, accurate and reliable for quality control of Xinxue granula.
