摘 要:目的探讨熊果酸在体外对单核细胞白血病细胞株THP1(humanacutemonocyticleukemiacellline)的增殖抑制和杀伤机制。方法采用MTT法和流式细胞术检测熊果酸对THP1细胞的增殖抑制和杀伤效应,hoechst33258荧光染色观察DNA片段化。结果熊果酸对THP-1细胞具有增殖抑制和杀伤效应,并呈浓度和时间依赖性。在熊果酸作用下THP-1细胞DNA受损,出现凋亡征象,G1期细胞数减少,细胞主要阻滞在S期。结论熊果酸在体外能诱导THP-1细胞凋亡。Objective To determine whether urselic acid(UA) can induce apoptosis in human acute monocytic leukemia(THP-1) cell line and its mechanism. Methods THP-1 cells were treated with UA. The proliferative inhibition was examined by MTF method. HOECHST 33258 fluorescent staining and flow cytometry were used to detect apoptosis. Results UA inhibited the proliferation of THP-1 cells. THP-1 cells treated with UA were induced to apoptosis. Apparent morphological index of apoptosis was observed. GI phase declined, cells were mainly arrested in S phase. Conclusion Apoptosis of THP-1 cell is the key mechanism for UA' s killing action in the treatment of cancer.
