摘 要:目的:观察三磷酸胞苷二钠(CTP)对缺血神经元修复、再生及结构重建的影响及其作用机制。方法:实验于2002-03/10在河北医科大学组胚教研室及河北医科大学第三医院中心实验室进行。选取SD大鼠60只,随机分为脑缺血自然恢复组、药物干预组和假手术对照组,每组20只。采用线栓法建立大脑中动脉脑缺血大鼠模型,药物干预组大鼠术后立即将三磷酸胞苷二钠(70mg/kg体质量)经1mL生理盐水溶解后给予腹腔注射,此后以相同剂量每日一次腹腔注射;自然恢复组及假手术组大鼠给予生理盐水1mL每日一次腹腔注射,分别于术后7、14、21及30d时各取5只大鼠处死,应用苏木精-伊红染色、免疫组化技术观察海马CA3区脑源性神经生长因子的表达,比较各组脑缺血后脑源性神经生长因子阳性细胞吸光度值。结果:实验选取的各组大鼠全部进入结果分析。①各组大鼠脑缺血后细胞形态观察结果:假手术组大鼠顶叶及海马区神经元排列整齐,形态完整,细胞数量较多,细胞核圆大且核仁明显。缺血组可见梗死灶,表现为正常结构消失,排列紊乱,细胞数量减少,胞核浓缩呈梭形或三角形,细胞质深染。②免疫组化染色及图像分析结果:光镜下观察,假手术组海马CA3区脑源性神经生长因子免疫组化染色阳性细胞神经元的胞浆内充满弥漫性的棕黄色颗粒,但其表达数量较少、染色较浅,自然恢复组脑源性神经生长因子表达与假手术组相比逐渐增多,第21天时达到高峰,并且免疫反应产物染色较深,部分神经元可见长的轴突,三磷酸胞苷二钠干预组脑源性神经生长因子阳性表达同自然恢复组相比均明显增加。③脑缺血后各组脑源性神经生长因子阳性细胞吸光度值的比较:与假手术组相比,自然恢复组脑源性神经生长因子吸光度值明显升高且差异有显著性(P<0.01),与自然恢复组相比,三磷酸胞苷二钠干预组各�AIM: To investigate the effect of sodium eytidine triphosphate (CTP) on the ischemic neuronal repair, regeneration and structural reconstruction and its mechanism. METHODS: This experiment was conducted at the Department of Histology and Embryology, Hebei Medical University and Central Laboratory, Third Hospital, Hebei Medical University from March 2002 to October 2002. Totally 60 SD rats were used and randomly divided into natural recovering group, CTP-treated group and sham-operation group, with 20 rats in each group. Unilateral cerebral ischemia was induced in rats by penna- fient occlusion of the distal middle cerebral artery. CTP (70 mg/kg body mass, mixed with 1 mL normal saline) was intraperitoneally injected into rats just after end ischemia and then the same dose of CTP was given to the rats in the CTP-treated group. Rats in the natural recovering group and sham-operation group were intraperitoneally given normal saline of 1 mL daily. Five rats were executed respectively at post-operative 7, 14, 21 and 30 days. Haematoxylin-eosin staining and immunohistochemicai technique were used to observe the expression of BDNF in the hippocampal subfields CA3 and compare the abserbance of BDNF positive cells after cerebral ischemia in each group. RESULTS: Rats used in each group all entered the stage of result analysis.(1) Observation of cellular morphology of the rats after cerebral ischemia: In the sham-operation group, parietal lobe and neurons in hippocampus arranged well, with complete morphology and the number of cell was numerous. Nuclei were large, spherical and obvious. Infarction was found in ischemia group, normal structure disappeared, arranged disorderly and the number of cells was decreased. Nuclei were also seen shuttle-shape or triangular, cytoplasm was dark stained. (2) Immunohistochemieal staining and image analysis results: Observation by the optical microscope, in sham-operation group, BDNF positive products were seen in hippocampal CA3 sections.
