摘 要:目的探讨雷公藤治疗系统性红斑狼疮(SLE)的免疫作用机制。方法采用电泳迁移率试验(EMSA)检测SLE患者雷公藤治疗前后和正常对照组外周血单一核细胞(PBMC)中NF-κB活性,同时对活动期SLE患者PBMC用雷公藤处理,比较其NF-κB活性变化。结果SLE患者和正常对照组均有一定量的NF-κB活化,但SLE患者NF-κB活性明显高于正常对照组(P<0.01),且活动期又明显高于非活动期(P<0.05)。经雷公藤处理后,活动期SLE患者NF-κB活性显著降低(P<0.01),与非活动期SLE患者相当(P>0.05),但仍明显高于正常对照组(P<0.05)。结论SLE的发病机制复杂,雷公藤可能仅在某些环节抑制了NF-κB的表达,在SLE治疗中发挥其免疫抑制作用。Objective To investigate the immunomechanism of Tripterygium Wilforddi Hook(TWH)in treatment of the patients with systemic lupus erythematosus (SLE). Methods The nuclear factor - κB ( NF - κB) activation in peripheral blood mononuclear cell (PBMC) from SLE patients and normal controls were detected by eleetrophoresis mobility shift assays (EMSA), Results Activation of NF - κB in PBMC were found in both SLE patients and normal controls, But NF-κB activation was increased significantly in PBMC from SLE patients, compared with that from normal controls ( P 〈 0. 01 ), and activation of NF-κB in active SLE group was higher than that of inactive SLE group( P 〈 0. 05 ). When the PBMC from active SLE patients were processed by TWH, the activation of NF-κB was decreased significantly ( P 〈 0.01 ), which was simular to that from inactive SLE patients, but still higher than that from normal controls ( P 〈 0, 05 ), Conclusion By down-regulating the of expression of NF-κB, TWH might exert the immunoimpression in the treatment of SLE.
