摘 要:目的:观察乙型肝炎病毒表面抗原中蛋白(MHBs)核酸疫苗在中国猕猴体内的体液和细胞免疫应答。方法:实验用中国猕猴实验组、对照组各2只,分别在第0、8、16、24、48周肌肉注射法接种核酸疫苗(pSW3891/MHBs)或对照载体(pSW3891)。ELISA法检测中国猕猴血清抗-HBs,CFSE-PI染色FCM(流式细胞术)法测定中国猕猴外周血淋巴细胞HBsAg特异性增殖反应。结果:实验组中国猕猴血清抗-HBs滴度在72周达到最高峰,最高滴度分别为1∶6400和1∶12800,对照组抗-HBs滴度始终保持在1∶50。实验组中国猕猴外周血淋巴细胞HBsAg特异性增殖反应明显增强,细胞分裂指数分别为18.64和14.70,而对照组细胞分裂指数分别为1.62和2.61。结论:建立了CFSE-PI染色流式细胞仪检测法来检测中国猕猴外周血淋巴细胞HBsAg抗原特异性增殖反应,方法创新,实用可靠。新型乙肝表面抗原中蛋白核酸疫苗pSW3891/MHBs在中国猕猴体内能产生良好的特异性体液和细胞免疫应答。Objective: To observe the humoral and cellular immune responses of rhesus macaques induced by a novel DNA vaccine encoding middle hepatitis B virus surface antigen(MHBs). Methods: Four rhesus macaques were divided into two groups. Each group received five i.m. doses of pSW3891/MHBs or the vector pSW3891 at 0, 8, 16, 24 and 48 weeks. Anti-HBs in sera Was tested by ELISA (enzyme linked immunosorbent assay). HBsAg specific T lymphocyte proliferative response in peripheral blood mononuclear cells (PBMCs) labeled with carboxyfluorescein diacetate succinimidyl ester (CFSE)-PI was analyzed by Flow Cytometry (FCM). Results: The rhesus macaques immunized with pSW3891/MHBs showed strong anti-HBs response (ending point 1 : 6400 and 1 : 12 800), compared with the controls (ending point 1 : 50). The proliferative response of the PBMCs of immunized rhesus macaques (cell division index 18.64 and 14.70) is stronger than the controls (cell division index 1.62 and 2,61). Conclusion: To assess specific T lymphocyte proliferation a flow cytometry assay using PBMCs labeled with CFSE-PI was established. A novel DNA vaccine (pSW3891/MHBs) encoding middle hepatitis B surface antigen can induce significantly humoral and cellular immune responses in rhesus macaques.
