乳腺癌RASSF1A基因表达及启动子区甲基化研究

摘　　要：目的:探讨抑癌基因RASSF1A启动子甲基化状态及其基因表达与散发性乳腺癌发生发展的关系。方法:采用甲基化特异性PCR(methylationspecificPCR)检测36例乳腺癌,12例癌旁组织,13例乳腺良性肿瘤组织中RASSF1A基因启动子甲基化状态,同时应用半定量RT-PCR(semi-quantitativereversetranscriptasePCR)检测其RASSF1A基因表达情况。结果:乳腺癌组织RASSF1A基因启动子区甲基化频率为61.1%,癌旁组织和良性乳腺肿瘤组织未发生甲基化;RASSF1A基因启动子甲基化与患者的年龄,病理组织学类型,临床分期,淋巴结转移无相关性。乳腺癌组织RASSF1A基因表达缺失率为33.3%,癌旁组织,良性肿瘤组织均表达RASSF1A;半定量RT-PCR结果表示乳腺癌组织RASSF1A基因表达量显著低于癌旁组织和良性肿瘤组织。结论:RASSF1A基因启动子甲基化可能是散发性乳腺癌中一类重要的分子改变,也是导致RASSF1A基因表达下降原因之一。Objective： To investigate the relationship between promoter methylation status and expression of tumor suppressor gene RASSFIA and tumorigenesis and development in diverging breast cancers. Methods： MSP （methylation specific PCR） was used to measure the promoter methylation status of 36 cases of breast cancers,12 adjacent normal tissues and 13 benign breast tumors. RASSF1A mRNA expression level was measured by semi-quantitative RT-PCR. Results： The frequency of promoter methylation of RASSFIA gene in breast cancers was 61.1%,while none of adjacent normal tissues and benign breast tumors was present with the promoter methylation, However,promoter methylation of RASSF1A gene in hreast cancers was not correlated with clinical parameters, such as age, histological type,TNM stageand lymph node metatasis. RASSF1A mRNA appeared in adjacent normal tissues, benign breast tumors. And the loss frequency was 33.3% in breast cancers. The mRNA expression level of RASSF1A was significantly lower in breast cancers than that in adjacent normal tissues, benign breast tumors. Conclusion： The promoter methylation of RASSF1A may be an important molecular change and may contribute to the low level of RASSF1A mRNA expression in diverging breast cancers.
