肾母细胞瘤组织中MVD、VEGF的表达及意义

摘　　要：目的检测肾母细胞瘤组织中血管内皮生长因子(VEGF)和微血管密度(M VD)的表达,并探讨二者与肾母细胞瘤病理分型的关系。方法手术切取50例肾母细胞瘤患儿的肿瘤组织,其中为组织结构良好型(FH型)40例,组织结构不良型(UH型)10例。采用免疫组织化学SABC染色法检测其M VD及VEGF表达。结果M VD值FH型为50.57±13.01,UH型为85.92±14.36,两型相比,差异有统计学意义(t=7.535,P<0.05)。VEGF表达率在FH型为65.00%,UH型中为60.00%,两型相比,差异无统计学意义(χ2=0.087,P>0.05)。VEGF阳性者M VD值为65.70±16.88,VEGF阴性者为48.18±18.13,两型相比差异有统计学意义(t=3.534,P<0.05)。结论肾母细胞瘤组织中VEGF阳性表达率和M VD较高。M VD与肾母细胞瘤病理分型有关,是判断肾母细胞瘤预后较为理想的指标。VEGF可能在促进肿瘤血管形成中起重要作用。[Objective] To investigate the expression of vascular endothelial growth factor （VEGF） and mierovesel density（MVD）,and study their relationships and the their role in the angiogenesis in nephroblastoma, reveal the relationships among them and the biogical characters of nephroblastoma. [Methods] 50 cases of HE staining nephroblastoma sections were affirmed and histopathologic types were classified. 40 cases had favorable histologic（FH） types, and 10 cases had unfavorable histologic （OH） types. The expression of MVD and VEGF were measured retrospectively with immunohistochemical staining （SABC）. [Results] The MVD in UH cases （85.92 ± 14.36） was significantly higher than that in FH cases （50. 57 ± 13. 01）, the difference had statisical signnificance（ P 〈0. 05）. The VEGF expression rate was 65.00% in FH type and 60. 00% in UH type, there was no statisical signnificance（ P 〉 0. 05）. The MVD in VEGF positive group （65. 70±16. 88） was higher than that of VEGF negative group （4.18 ± 18. 13）, the difference had statisical signnificnoce （ P 〈 0. 05） . [Conclusion] The MVD is an indicator of the prognosis of patients with nephroblastoma. The VEGF expression has no association with the histological type, but plays an important role in the invasion and metastasis of nephroblastoma through angiogenesis and cell proliferation.
