雌激素对骨质疏松大鼠骨胶原的影响

摘　　要：目的观察雌激素对卵巢切除所致骨质疏松症大鼠骨胶原的影响。方法7月龄未交配雌性SD大鼠30只,随机平分为OVX组(仅予卵巢切除)和雌激素组(予卵巢切除和雌激素治疗)。12周后处死大鼠,测定其第5腰椎羟脯氨酸(HYP)含量,同时应用M asson三色法行第4腰椎椎体切片染色,光镜下观察胶原纤维形态学的改变。结果卵巢切除12周后,OVX组大鼠第5腰椎HYP含量为(0.182±0.022)m g/g,镜下见胶原纤维排列稀疏紊乱,变细甚至断裂溶解,连续性差。雌激素组HYP含量(0.202±0.017)m g/g,镜下见胶原纤维排列规则,走向清晰,连续性好,胶原纤维粗壮,呈致密板层样。两组HYP含量相比差异有统计学意义(P<0.05)。结论卵巢切除致骨质疏松,骨胶原含量下降。雌激素能增加骨胶原含量,为骨矿化提供框架核心。[Objective] To find the effects of estrogen on the bone collagen in the osteoporosis rats. [Method] Normal 30 null mating seven-month old femeale SD rats were divided randomly into two groups, each gnoup included 15 rats. The OVX group was ovariectomized; the estrogen group was treated with dihydroxyestrin after ovariectomy. Executed the rats after 12 weeks, determined the HYP of the 5th lumbar vertebra, and observed the collagen morphology of the 4th lumbar vertebra using the microscope after the masson triad color dyeing. [Result] HYP in OVX group was （0. 182±0. 022） mg/g. The bone collagen was incompact, thin, collapse and poor succession. HYP in the estrogen group was （0. 202±0. 017） mg/g. The bone collagen was regular, compact, and closed succession. [Conclusion] Ovariectomy can lead to osteoporosis, and there is a decrease on the bone collagen in the ovariectomized rats. The estrogen can increase the content of the bone collagen which provide the core for the bone mineralization.
