摘 要:目的在成功培养、纯化体外鸡胚额骨成骨细胞的基础上,探讨不同剂量重组鸡IGFⅠ(rcIGFⅠ)对成骨细胞骨相关基因(COLⅠ、OPG、IGFⅠR)表达的影响。方法应用酶消化法(0.1%Ⅱ型胶原酶)获取鸡胚额骨成骨细胞群并鉴定。取培养2d的2代成骨细胞,在rcIGFⅠ为0ngml、50ngml、100ngml的浓度中进行培养,采用RTPCR方法检测基因COLⅠ、OPG、IGFⅠR的相对表达情况,并用βactin作为内参照予以矫正。结果rcIGFⅠ可以在转录水平上增加鸡胚额骨成骨细胞骨相关基因的表达,并呈明显的剂量依赖关系。结论IGFⅠ促进成骨细胞的增殖、分化的作用可能是通过增加骨相关基因的表达水平来实现的。Objective To study the effect of recombinant chicken IGF- Ⅰ on the bone-related gene expression based on effective method of isolating and purifying osteoblastic cells from calvariae of embryonic chicken. Methods The osteoblastic cells were harvested by enzyme digestion (0. 1% ellogenase Ⅱ )from bone chips isolated from ealvariae of about 15-day embryonic chicken and idemified with AKP staining. Then the osteoblasts of second generation planted for 2 days were cultured in medium with different concentrations of rclGF- Ⅰ (0 ng/ml,50 ng/ml, 100 ng/ml). The abundance of COL- Ⅰ ,OPG,IGF- Ⅰ R mRNA in osteablastic cells were measured by RT- CPR with β-actin as internal standard. Results The expression of COL- Ⅰ ,OPG,IGF- Ⅰ R genes increased at the levels of transcription dose-dependently. Conclusions IGF-Ⅰ might induce the proliferation and differentiation of osteoblasts by increasing the expression of bone-related genes.
