摘　　要：VP41蛋白质是对虾白斑综合症病毒的一种结构蛋白,该蛋白质由病毒基因组上的wsv242开放阅读框(ORF)所编码,分子量为41 ku.根据vp41基因的序列,通过PCR扩增获得了该全长基因,将其克隆在载体pET-His上,以E.coliBL21为宿主菌,成功表达并纯化了含His标记的目的蛋白VP41,并制备了特异性鼠抗血清.当纯化的病毒粒子经去污剂处理后,VP41蛋白质被发现完全存在于病毒WSSV囊膜部分,Western blot杂交实验也证实该结果,表明VP41蛋白质是该病毒的一个囊膜蛋白质.通过Far-Western方法还发现VP41具有蛋白聚合作用.White Spot Syndrome Virus （WSSV） is a major virulent pathogen in shrimp aquaculture,and can also infect most species of crustacean. First appearing in the 1990s in Taiwan,WSSV has quickly spread to Southeast Asia,the Indian continent and Central-and Latin-American, causing catastrophic economic losses. We described herein the characterization of VP41 ,one of major structural proteins of WSSV. It was a 41 ku novel envelop protein from WSSV, being characterized by SDS-PAGE. The gene was expressed in Escherichia coil and purified,then was used to prepare specific antibody. Meanwhile,it was found taking on polymer. When purified virions were treated with Triton X-100 detergent,VP41 was found to present exclusively in the envelope fraction,which was also confirmed by Western blot analysis. The result indicated that VP41 behaved as an envelope protein.
