摘 要:目的:探讨微囊化牛嗜铬细胞蛛网膜下腔移植的镇痛作用,分析海藻酸钠-多聚赖氨酸-海藻酸钠微囊的免疫隔离效果。方法:实验于2003-08/2004-09在郑州大学医学院解剖实验室完成。①选取清洁级成年SD大鼠96只,随机数字表法分为:空囊组、微囊化细胞组、裸细胞组,32只/组。②空囊组向大鼠蛛网膜下腔植入40μL含空囊的DMEM液,每只植入600~800个海藻酸钠-多聚赖氨酸-海藻酸钠空囊;微囊化细胞组向大鼠蛛网膜下腔植入40μL海藻酸钠-多聚赖氨酸-海藻酸钠-牛嗜铬细胞悬液,含细胞5×106个;裸细胞组向大鼠蛛网膜下腔植入40μL牛嗜铬细胞悬液,含细胞5×106个。③分别于细胞移植术后2,4,6,8周测定各组大鼠脑脊液灌流液中儿茶酚胺含量以及脊髓后角fos蛋白表达的变化,各时间点8只/组。结果:实验选取清洁级成年SD大鼠96只,全部进入结果分析。①海藻酸钠-多聚赖氨酸-海藻酸钠微囊化牛嗜铬细胞的形态:光镜下,包裹嗜铬细胞的微囊呈规则的圆球形,表面光滑,直径为150~250μm。牛嗜铬细胞散在分布于微囊内,细胞呈圆形,清晰可见。空囊半透明状,表面光滑。②移植术后不同时间各组脑脊液灌流液中儿茶酚胺的含量检测结果:微囊化细胞组和裸细胞组大鼠脑脊液灌流液中儿茶酚胺的含量在移植术后2,4,6,8周均高于空囊组(P<0.05)。微囊化细胞组移植术后2周与裸细胞组基本相似(P>0.05),其余各时间点均高于裸细胞组(P<0.01)。③移植术后8周各组大鼠脊髓内fos蛋白表达的变化:大鼠注射甲醛侧脊髓灰质后角内阳性神经元个数微囊化细胞组、裸细胞组均显著低于空囊组[(39.96±4.46),(50.62±3.29),(59.70±6.93),P<0.05]。同时,微囊化细胞组脊髓组织中阳性神经元个数明显低于裸细胞组(P<0.05)。结论:微囊化牛嗜铬细胞移植可以提高脑脊液中儿茶酚胺的含量,同时抑制甲醛致痛大鼠脊髓后角fos蛋白的�AIM: To study the analgesic mechanism of transplantation of microencapsulated bovine chromaffin cells (BCCs) into the spinal subarachnoid, so as to observe the immune isolation effect of Alginic acid-Poly-l:lysine-Alginic acid (APA) microcapsule. METHODS: The experiment was conducted in the dissecting laboratory of Medical College of Zhengzhou University between August 2003 and September 2004. ①Totally 96 clean grade adult Sprague-Dawley rats were randomly divided into APA group, APA-BCC group and BCC group with 32 rats in each group. ②The rats of APA group were injected with 40 μL DMEM including .600-800 APA empty micrecapsules into the spinal subarachnoid space; The rats of the APA-BCC were injected with 40μL APABCCs suspension containing 5×10^5 cells into the spinal subarachnoid space; The rats of the BCC group were injected with 40μL BCCs suspen sion containing 5×10^5 cells into the spinal subarachnoid space. ③The concentration of catecholamine in rat cerebrospinal fluid and the expression of. C-fos gene in spinal horn cord were detected at 2, 4, 6, and 8 weeks after operation with 8 rats at each time point. RESULTS: The 96 SD rats were all involved in the result analysis. ①Appearance of APA-BCC: Under the electronic microscope, the microcapsules encapsulated chromaffin cells showed regular round, silky surface, a 150-250μm in diameter. The BCCs were scattered in microcapsules, and were round and clear. The empty "microcapsules were translucent with silky surface. ②Concentration of catecholamine in rat cere brospinal fluid at different time points after transplantation: The concentration of the catecholamine in the APA-BCC group and BCC group was higher than that in the APA group at each time point after transplantation (P 〈 0.05). ,The APA-BCC group was similar to that of the BCC group at the 2nd week after operation (P 〉 0.05), while higher than the BCC group at the other time points (P 〈 0.01). ③Changes of expression of C-fos gene in
