水稻Os WRKY10与GFP融合基因的烟草转化及亚细胞定位观察

摘　　要：OsWRKY10是一个新的WRKY类转录因子。该项研究构建了OsWRKY10基因与绿色荧光蛋白基因融合的增强表达载体pCamU-OsWRKY10-GFP。将载体转入根瘤农杆菌EHA105中,叶盘转化法转化烟草。转化植株经PCR鉴定呈阳性。通过荧光显微镜可检测到绿色荧光蛋白在根细胞的核中表达,并且OsWRKY10基因定位在细胞核。OsWRKYIO is a new WRKY transcription factor. In this experiment, fusion gene of OsWRKYIO and green fluorescent protein （GFP） were inserted into a pC, ainU vector for constructing the p]ant expressing vectorpCamU-OsWRKY10 - GFP. Then,the recombinant vector was transformed into Nicotina tabaccum L.by A. tumefanciens -mediated method. Transgenic tobacco plants were detected by PCR method. The result showed that the gene was integrated into the genomes of tobaeco plants. Green fluorescence was visualized under blue light excitation in root cells of transgenic tobacco plants by fluorescence microscopy. The study also revealed that OsWRKYIO gene was exclusively localized in the nuclear, which was suggesting that it was a nuclear regulator.
