大鼠睾丸特异表达新基因Lrrc18的克隆及表达谱分析

摘  要:运用数据库消减杂交(Digital Differential Display,DDD)分析方法,从大鼠睾丸组织中分离出了一个新基因-Lrrc18(GenBank登录号:BC081908).该基因定位于大鼠染色体16p16区带,gDNA在染色体上跨度40900bp,含6个外显子,编码一个含256个氨基酸的蛋白,带有4个LRR(leucire-rich repeat domain)结构域.Northern杂交结果显示Lrrc18只在睾丸中表达.对出生后3~56 d不同时期的大鼠睾丸的Lrrc18基因的表达进行检测,结果表明在大鼠出生后21 d时开始在睾丸组织中可检测到Lrrc18的mRNA表达,以后其表达量逐步上升,到42 d时达到最大值.该结果提示:Lrrc18基因在精子的发生过程中可能起重要作用.A novel testis-specific gene, termed Lrrc 18 (GenBank accession number: BC081908), was identified by digital differential display. Lrrc 18 gene is located in chromosome 16p16 region, spans approximately 40 900 bp, and is organized into 6 exons. The entire open reading frame encodes 256 amino acid residues which has 4 LRR ( leucine-rich repeats) domains. Northern blot hybridization and RT-PCR results showed that Lrrc 18 mRNA was specifically expressed in adult rat testis. Expression levels of Lrrc 18 in rat testis from postnatal day 3 to day 56 were examined, the results showed that low-level Lrrc 18 mRNA was first detected on day 21, then the expression of Lrrcl8 mRNA increased gradually with the development of the testis of rat, and it reached the peak value on day 42. The time-dependent expression pattern of Lrrc 18 in postnatal rat testes suggested that Lrrc 18 gene might be involved in the regulation of spennatogenesis and sperm maturation.
