摘 要:目的探讨扁桃酸在小鼠体内抗弓形虫速殖子的作用机制。方法给予弓形虫感染的小鼠口服扁桃酸200 mg/kg,2次/d,然后在不同时间取腹水及肝脏制作电镜标本,观察弓形虫的超微结构变化。结果腹水中游离弓形虫速殖子和肝组织内弓形虫速殖子的超微结构均发生明显改变,细胞膜迂曲破裂,线粒体、内质网扩张破裂,棒状体肿胀破裂,致密颗粒减少或消失,核膜迂曲破裂,核固缩。扁桃酸实验组小鼠肝细胞的受损程度明显轻于阳性对照组和乙胺嘧啶药物对照组的肝细胞改变。结论通过形态学观察,发现扁桃酸的作用机制主要表现在其对虫体细胞膜、入侵细胞器及细胞核等结构的破坏,导致虫体崩解或抑制虫体的繁殖。Objective To study the mechanism of mandelic acid against toxoplasmic tachyzoite in mice. Methods Mandelic acid (200 mg/kg, twice daily) was orally given to the infected mice; and the ascites and liver tissue were taken at different time to make electromicroscopic sample in order to observe the microstructure changes of toxoplasmic tachyzoite. Results Microstrctures of toxoplasmic tachyzoite, either in ascites or in liver tissue, were obviously changed. It mainly showed that., cell membrane was tortuous and broken, mitochondrial and endplasic reticulum were enlarged and broken, rhoptry was swollen and broken, dense granules decreased or disappeared, and nucleolus was contracted. Degree of liver damage in mandelic acid group was lighter than that in pyrimethine controlled group and positive group. Conclusion The mechanism of mandelic acid against toxoplasmic tachyzoite in morphology mainly shows that mandelic acid destroys the parasite cell membrane, invasive organs and nucleus et al, and thus causes worm destruction or inhibits the worm reproduction.
