清利固精颗粒对系膜增殖性肾炎IL-1β及TGF-β1的影响

摘　　要：目的:探讨清利固精颗粒(主药:地骨皮、芡实、土茯苓)对大鼠抗Thy1系膜增殖性肾炎(MsPGN)IL1β及TGFβ1的影响。方法:本研究应用大鼠抗Thy1系膜增殖性肾炎(MsPGN)动物模型,将大鼠随机分为4组:正常对照组、模型组、肾炎康复片组、清利固精组,每组8只大鼠。各实验组于第7天处死大鼠,检测肾小球培养上清IL1β含量,检测肾组织TGF-β1含量。结果:模型组大鼠肾小球培养上清IL1β含量及肾组织TGFβ1含量比正常对照组明显升高(P<0.01);与模型组相比,清利固精组肾小球培养上清IL1β含量及肾组织TGFβ1含量明显下降(P<0.01);清利固精颗粒降低肾组织TGF-β1含量疗效优于肾炎康复片(P<0.05)。结论:清利固精颗粒能降低肾炎大鼠肾小球培养上清IL-1β水平和肾组织TGF-β1含量,抑制细胞增殖及细胞外基质增加,可能是其治疗MsPGN作用机制之一。Objective： To investigate the effect of QGG （main ingredients： Lycium Chinese Mill, Euryaie ferux Salisb, Smilax glabra Roxb） on IL-1β and TGF-β1 with MSPGN. The model was induced by repeated injection of anti-Thy1 antiserum. Methods： 32 rats were randomly divided into four groups, the contrul group, the MSPGN group, the Shenyan Kangfu Tablet （SKT） -treated group and the QGG-treated group.Kidney were taken on day 7 to measure The level of IL-1β in cultured Mesangial cells with RIA and the expression of TGF-β1 with immuno-histochemistry. Results： the level of IL-1β in cultured Mesangiai cells and the expression of TGF-β1 increase in the MSPGN rats as compared with the control rats （ P 〈 0.01 ） .The level of IL-1β in cultured Mesangial cells and the expression of TGF-β1 decrease in the QGG-treated rats as compared with the MsPGN rats （ P 〈 0.01 ） and the effect of decreasing the expression of TGF-β1 in the QGG-treatcd rats was better than the SKT-treated rats （ P 〈 0.05） . Conclusion： The level of IL- 1β in cultured Mesangial cells and the expression of TGF-β1 in kidney tissue of the MsPGN rats was decreased effectively when treated by QGG. QGG can inhibit mesangiai cell and extra cellullarmatrix proliferation. QGG will be available in treating MsPGN and prevent the disease worsening.
