妊娠期糖尿病孕妇外周血单个核细胞TNF-α基因表达水平的实验性研究

摘　　要：目的:观察妊娠期糖尿病(GDM)患者外周血单个核细胞(PBM C)中肿瘤坏死因子-α(TNF-α)基因转录和表达的水平,探讨其在GDM发病过程中的意义。方法:选取GDM、正常妊娠和健康非妊娠组各30例,采用EL ISA法检测空腹血清中的TNF-α水平。收集研究对象外周血单个核细胞(PBM C),采用半定量逆转录-聚合酶链反应(RT-PCR)和免疫印记分析(W estern b lot)技术测定实验对象外周血单个核细胞TNF-αmRNA转录和蛋白质表达的水平。结果:GDM组、正常妊娠组空腹血清TNF-α水平显著高于健康非妊娠组,而GDM组显著高于正常妊娠组。三组间的外周血单个核细胞TNF-αmRNA转录水平、TNF-α蛋白的相对含量无显著性差异。结论:TNF-α可能参与了GDM的发生发展过程,但是PBM C可能不是GDM时TNF-α增加的来源。Objective： To study the expression changes of tumor necrosis factor -alpha （TNF-α） mRNA and protein in patients with gestational diabetes mellitus （GDM） by Peripheral blood mononuclear cells （PBMC）, Methods： 30 women with GDM, 30 health pregnancy women （NGT） and 30 health non-pregnancy women were studied. Serum TNF-α were measured by Elisa. Peripheral blood mononuclear cells （PBMC） were isolated from plasma,then levels of TNF-α mRNA and protein of PBMC were determined by RT-PCR and Western blot in 3 groups. Results ： GDM and NGT pregnancy women showed higher level of TNF-α than non-pregnancy women. GDM group showed higher level of TNF-α than NGT group. There were no difference among three groups in level of TNF-α mRNA and protein of PBMC. Conclusion： The expression of tumor necrosis factor-alpha in patients with gestational diabetes mellitus was up regulated and PBMC may be not the source of TNF-α. It suggested that TNF-α did not interact in the development of GDM.
