圣和散逆转胃癌SGC7901/VCR细胞多药耐药的研究

摘　　要：目的探讨中药圣和散逆转胃癌耐药细胞SGC7901/VCR多药耐药的机制。方法圣和散(浓度2.5、5和10mg/ml)处理胃癌耐药细胞SGC7901/VCR,用流式细胞技术检测P-糖蛋白(P-gp)表达,免疫组织化学SABC法及图像分析技术检测B细胞淋巴瘤/白血病基因-2(Bcl-2)。结果实验组SGC7901/VCR细胞Bcl-2表达A值(121.34±0.16)明显高于对照组(104.37±0.089)(P<0.05),Bcl-2表达减少,P-gp表达也明显降低。结论圣和散能逆转胃癌耐药细胞SGC7901/VCR多药耐药,其作用可能与调节Bcl-2和P-gp表达有关。Objective：To explore the mechanism of Shenghe Powder（SHP） in reversion of multidrug resistenee of SGC7901/VCR cells of gastric cancer. Methods：Gastric cancer drug- resistance cells SGC7901/VCR were treated with SHP at concentrations of 2.5,5.0 and 10mg/ml. The expression of P-glyeoprotein（P-gp） was detected with floweytometry and B-cell lymphoma/leukemia-2（Bcl- 2） protein was examined by immunohistochemieal SABC method and imaging analysis. Results：In SHP treated group ,A value of Bcl-2 in SGC7901/VCR cells was significantly higher than that in the control group（P（0. 05）,and the expression of Bcl-2 and P-gp was significantly down-regulated. Conclusion ：SHP can reverse the multidrug resistance of SGC7901/VCR ,which is possibly related with the changes of the expressions of Bcl-2 and P-gp.
