摘  要:目的:探讨中药助孕胶囊(ZYJN)对多囊卵巢(PCO)大鼠的治疗作用并分析其可能的作用机制。方法:采用皮下埋植左旋18-甲基炔诺酮硅胶棒的方法制作动物模型,将大鼠随机分为模型组、助孕胶囊组、克罗米酚组、联合用药(助孕胶囊+克罗米酚)组及对照组,运用放免法测定血性激素水平的变化,光镜观察卵巢、卵泡发育情况,免疫组化法检测卵巢组织胰岛素样生长因-1的表达水平。结果:助孕胶囊可使PCO大鼠的颗粒细胞层增厚,卵泡膜细胞层变薄,血清T、LH水平均明显下降,并下调卵巢组织胰岛素样生长因子-1的表达。结论:助孕胶囊能够通过调整内分泌及影响卵巢胰岛素样生长因子-I的表达,在中枢及卵巢局部协调发挥作用,从而调整性腺功能,促进卵泡的发育成熟及排卵。Objective: To investigate the effects of zhuyunjiaonang (ZYJN) on rats with polycystic ovary and to analyze the possible mechanism. Methods: Polycystic ovary rat models were established by imbedded L18-methylnorethindrone subcutaneously. The rats were divided into the model group, ZYJN group, chloramiphene group, associated medication group (ZYJN and chloramiphene) and vehicle group. The levels of serum gonadal hormone were detected by radioimmunoassay. The effects on ovaries and ovarial follicular development were observed by light microscope. Using the immunohistochemical staining technique detected the expressed level of insulin-like growth factor-1 (IGF-1) Results: ZYJN could increase the thickness of granular cell layer ovary and decrease the thickness of theca cell layer, ZYJN greatly decreased the level of T and LH in rat serum as well as the express of insulin-like grown factor-1 (IGF-1) in rat ovary. Conclusion : ZYJN could play a role coherently in center and ovary by regulating the endocrine function of rat and affecting the expression of the IGF-1 in rat ovary thus adjusting genitical gland and promoting the mature and ovulation of follicular.
