布洛芬对创伤大鼠淋巴细胞内环核苷酸水平白细胞介素─2生成及NK细胞活性的影响

摘　　要：目的：研究腹部创伤大鼠免疫功能变化及布洛芬对创伤大鼠的免疫调节作用．方法：白细胞介素－２（ＩＬ－２）诱生，采用ＣＴＬＬ细胞增殖法．ＮＫ细胞活性检测，采用乳酸脱氢酶释放法．淋巴细胞内ｃＡＭＰ与ｃＧＭＰ浓度测定，采用放射免疫法．结果：腹部创伤大鼠脾淋巴细胞ＩＬ－２生成能力明显降低，脾脏ＮＫ细胞活性显著受到抑制，脾淋巴细胞内ｃＡＭＰ浓度下降，ｃＧＭＰ水平升高．应用布洛芬治疗能够增强创伤大鼠ＩＬ－２生成能力，恢复ＮＫ细胞活性，淋巴细胞内ｃＡＭＰ浓度回升，ｃＧＭＰ水平下降．结论：布洛芬能够明显改善创伤大鼠的免疫功能。Aim:TostudythechangesofimmunefunctionsandimmunomodulatoryefectsofIbuprofeninratswithabdominaltrauma.Methods:Assayofinterleukin2(IL-2)wasperformedbyusingthemethodofCTLLcelprolifera-tion.MeasurementofNKactivitywasconductedaccordingtothemethodoflactatedehydrogenaserelease.IntracelularcAMPandcGMPweredetectedbyradioimmunoassaymethod.Results:IL-2productionandintracelularcAMPcontentofsplenocytesinratswithabdominaltraumaweresignificantlydecreasedanditsNKactivitywasalsodepressedobvi-ously.ThelevelofintracelularcGMPwasmarkedlyincreased.Ibuprofen,usedbyintraperitonealinjection,couldgreatlypromptIL-2productionandintracelularcAMPlevel,recoverNKactivityanddiminishintracelularcGMPcon-tentinratswithabdominaltrauma.Conclusion:Ibuprofencouldclearlyimproveimmunefunctionsandmayincreasetheabilityagainstinfectioninratswithtrauma.
