摘  要:为了有效地研究胚泡附植的分子机制,本研究建立了一套适合兔子宫内膜蛋白分析的双向电泳技术体系,包括蛋白质含量测定、第1向IEF-PAGE和第2向SDS-PAGE的凝胶配方及电泳参数的选择、电极溶液的选择,并运用该技术体系分析了未孕兔和受孕2、4、6、9 d兔子宫内膜蛋白。结果表明,在给定的时期内,兔子宫内膜蛋白质的含量有变化,但差异不显著;用所建立的双向电泳体系分析子宫内膜蛋白质后发现,双向电泳图谱的重复性好,蛋白质点的分辨率高,共同蛋白质点多,差异蛋白质点少。In order to investigate the mechanism of embryo implantation effectively,a kind of two-dimensional electrophoresis system for rabbit endometrium analysis was established including protein content measurement,gel ingredients for IEF and SDS-PAGE,selection of electrophoresis parameters and electrode buffer using for the rabbit endometrium. Proteins in the nonpregnant rabbit endometrium and the 2th,4th, 6th,gth pregnant rabbit endometrium were also analyzed via the established system, The results demonstrated that there was some extent of variation of the endometrium total, protein content ,but the difference was not significant. Two-dimenslonal electrophoresis analysis of the given period endometrium proteins showed that the repeating of each diagram was excellent and the protein differentiation was also good. Moreover ,there were many common protein spots along with a few different protein spots for each given period diagram.
