Ru486对胎鼠肺Ⅱ型上皮细胞的影响

摘　　要：为研究糖皮质激素的可能作用位点，以分离培养的肺Ⅱ型上皮细胞为主要材料，通过板层小体染色的方法，观察Ｒｕ４８６对该细胞的影响。结果显示，无血清培液中Ｒｕ４８６１０^（－４）ｍｏｌ／Ｌ至培养第１ｄ或１０^（－５）ｍｏｌ／Ｌ培养至第３ｄ时均使Ⅱ型细胞出现坏死现象，但加入地塞米松（ＤＥＸ）１０^（－６）ｍｏｌ／Ｌ４ｈ后再用Ｒｕ４８６则坏死现象消失。由此从形态学角度证实了肺Ⅱ型上皮细胞具有糖皮质激素受体。Ｒｕ４８６是通过阻断该受体发挥作用的。PURPOSE To study the possible site of the glucocorticoid's effects. METHODS By the culture of purified fetal rat lung type Ⅱepithelial cells and the staining method of lamellar bodies,the effect of Ru486 on type Ⅱ cells were examined. RESULTS The results showed that the growth of cells could be inhibited by different concentration of Ru486.At the concentration of Ru486 10-4mol/ L or 10-5mol/L, the necrolysis of type Ⅱ cells happened on the 1st day and 3rd day respectively. But when DEX 10-6 mol/L was added 4h before Ru486 10-5mol/L, the effect of inhibition could be removed. CONCLUSIONS It suggested that there are glucocorticoid receptors on the type Ⅱ cells and such effect of Ru486 was through the receptors.
