猪带绦虫六钩蚴45W-4B重组蛋白的免疫原性研究

摘　　要：目的观察猪带绦虫六钩蚴45 W-4B抗原对仔猪的免疫保护作用。方法用IPTG诱导表达融合蛋白GST-45W-4B,用GST柱进行纯化,以Montan ide ISA 206为佐剂制成疫苗免疫仔猪。间接ELISA检测仔猪血清抗45W-4B IgG抗体水平,MTT法进行外周血淋巴细胞殖试验。免疫1个月后全部实验组经口攻击感染25000枚/头猪带绦虫虫卵,攻虫3个月后剖检,计算减虫率及保护率。结果免疫组注射疫苗的第2周起,抗体为阳性,45W-4B组持续升高至第8周,淋巴细胞增殖明显高于未免疫感染组。用该抗原制备的疫苗获得了95%的减虫率和33%的保护率。结论该抗原对猪囊尾蚴病具有很好的免疫保护作用。Objective To observe the immune protection induced by 45W- 4B antigen of Taenia solium oncosphere in pigs. Methods GST- 45W- 4B fusion protein was induced by IPTG and purified by Bulk GST Purification Module,the protein with Montanide ISA 206 was made into vaccine and injected into neck muscle of 2-month-old pigs. Anti 45W-4B antibody IgG in serum was screened by indirect ELISA. The blood lymphocytes proliferatior were evaluated by MTr methods. 1 month after the first immunization, all pigs were infected by mouth with 25000/ pig worms of Taenia solium. 3 months after challenge, all pigs were dissected, and the parasite decrease rates and protection rates were calculated. Results Antibody was detectable 2 weeks after the first immunization and 45W- 4B group continued to increase until week 8, the blood lymphocytes proliferation of the 45W- 4B vauine group was higher than blank infected group. The parasite decrease rate of GST- 45W- 4B vaccine was 95% and protection rate was 33%. Conclusion 45W- 4B antigen of Taenia solium oncosphere induces good immune protection in pigs.
