三步序贯法对激素干预后哮喘大鼠模型血清IL-4、IFN-γ含量及其肺组织mRNA表达的影响

摘  要:目的观察三步序贯法对激素干预后哮喘大鼠模型血清IL-4、IFN-γ含量及其肺组织mRNA表达的影响。方法将SD大鼠随机分为正常对照组、哮喘模型组、激素干预组、三步序贯组、第一步方组、第二步方组、第三步方组、普米克令舒组。制备激素干预后哮喘大鼠模型,于第3周起开始撤减激素用量,按照每周地塞米松0.1mg/kg速度递减,直至第7周完全撤除。采用ELISA法检测血清中IL-4和IFN-γ含量,用原位杂交技术检测各组大鼠肺组织中IL-4mRNA和IFN-γmRNA的表达变化。结果与哮喘模型组和激素干预组比较,三步序贯组IL-4的血中含量和肺组织中mR-NA的表达在各个时段均有显著性降低(P<0.01),而IFN-γ的血中含量和肺组织中mRNA的表达在各个时段均有显著性升高(P<0.01),与普米克令舒组比较,在第2周末,二者无统计学意义(P>0.05);在第7、9周末皆有极显著性差异(P<0.01)。结论三步序贯法可减少激素干预后哮喘大鼠模型IL-4血中含量及其在肺组织中的mRNA表达,提高IFN-γ血中含量及其在肺组织中mR-NA表达,调节IL-4/IFN-γ的失衡,减轻气道炎症的作用。Objective To observe the influence of 3-step sequential therapy on serum interleukin-4 (IL- 4), IFN-γ and lung mRNA expression in rat asthma model interfered with hormone. Methods To divide SD rats randomly into the normal control group (group 1 ), asthma model group (group 2 ), hormone interfered group ( group 3 ), 3-step sequential therapy group ( group 4), first-step group ( group 5 ), second-step group ( group 6), third-step group ( group 7 ) and Pulmicort Respules group ( group 8 ). After the establishment of rat asthma model interfered with hormone and on third week the hormone was withdrawn successively at the dose of 0. 1 mg/kg per week, and until 7^th week the hormone was withdrawn completely. The contents of serum IL-4 and IFN-γ were detected with ELISA method, and the expressions of IL-4 mRNA and IFN-γ mRNA in the lung tissue were examined by in situ hybridization in different groups. Results The contents of serum IL-4 and its lung mRNA expression were decreased significantly in different phases ( P 〈 0. 01 ), and the content of serum IFN-γ and its lung mRNA expression were increased significantly in different phases (P 〈0.01 ) in group 4 compared with that of group 2 and group 3. There was no statistical significance in the comparison between group 4 and group 8 (P〉0. 05 ) at the end of 2nd week, but a great significant difference at the end of 7^th and 92 weeks (P〈0. 01 ). Conchmion The 3-step sequential therapy can decrease, in rat asthma model interfered with hormone, the content of serum IL-4 and its lung mRNA expression, increase the content of IFN-γ and its lung mRNA expression, regulate the imbalance of IL-4 and IFN-γ , and relieve the inflammation of respiratory tract.
