摘 要:目的探讨穴位敷贴治疗哮喘的机理。方法将40只豚鼠随机分为正常对照组、哮喘模型组、穴位敷贴治疗组、地塞米松治疗组,每组10只。处理结束后采用TUNEL法检测支气管肺泡组织EOS凋亡;免疫组化法检测凋亡相关基因Fas、Fas配体(FasL)、bcl-2表达。结果模型组豚鼠支气管肺组织EOS凋亡指数及Fas、FasL、bcl-2表达与正常对照组比较差异均无显著性。穴位敷贴组和地塞米松组EOS凋亡指数及Fas、FasL表达明显高于模型组(P<0.01);地塞米松组和穴位敷贴组之间有显著性差异,前者明显高于后者(P<0.01)。穴位敷贴组bcl-2表达低于模型组(P<0·05);地塞米松组和模型组之间差异无显著性。结论穴位敷贴可以上调支气管肺泡组织促凋亡基因Fas及其配体FasL表达、下调凋亡抑制基因bcl-2表达、促进哮喘主要炎性细胞EOS凋亡。这可能是其治疗哮喘的作用机理之一。Objective To investigate the mechanism of acupoint dressing therapy for treating asthma.. Methods To divide randomly 40 guinea pigs into the normal control group ( group 1 ), asthma model group (group 2), acupoint dressing group (group 3) and dexamethasone group (group 4, each n = 10). The apoptosis of bronchoalvealar eosinocytes (EOS) was detected by TUNEL, and the expressions of Fas, FasL and bcl-2 genes were detected by immunohistochemical assay. Result The difference was not significant ( P〉0.05 ) in EOS apoptosis index, expressions of Fas, FasL and bcl-2 genes between group 2 and group 1. The EOS apoptosis index, expressions of Fas and FasL in group 3 and group 4 were higher significantly than that in group 2 (P〈0.01 ). There was a significant difference between group 4 and group 3. The index and expressions in group 4 was higher significantly than that of group 3 (P〈0. 01 ). The bcl-2 expression in group 3 was lower than that of group 2 (P〈0.05). The difference was not significant between group 4 and group 2 (P〉0.05). Conclusion The therapy of acupoint dressing can increase the expressions of Fas and FasL genes in bronchoalvealar tissue, decrease bcl-2 expression and promote the apoptosis of EOS, which may be its one of the mechanism for treating asthma.
