Ⅰ型胶原及核因子-κB与动脉粥样斑块的关系

摘　　要：目的:探讨人颈动脉粥样硬化(AS)时血脂成分,Ⅰ型胶原mRNA及其蛋白与NF-κBmRNA及其蛋白在人颈AS病变中的表达及临床意义。方法:对18例行颈动脉造影确诊有颈AS病变的患者,测定血脂成分,且行颈动脉内膜剥脱术,取出粥样斑块送病检;另取6例意外死亡(排除心血管疾病)的颈动脉标本作为对照;石蜡包埋制成切片。采用免疫组化及原位杂交法染色,检测Ⅰ型胶原mRNA及其蛋白与NF-κBmRNA及其蛋白在AS斑块中表达,并通过CMIAS真彩色医学图像分析系统测出表达的定量A值,进行2组间比较。结果:人颈AS病变中Ⅰ型胶原、NF-κB蛋白及Ⅰ型胶原mRNA、NF-κBmRNA表达定量A值,分别为:0.27±0.08、0.34±0.10及0.49±0.12、0.58±0.15,显著高于对照组(0.10±0.01、0.10±0.02及0.17±0.02、0.18±0.02)的定量表达(P<0·05)。结论:Ⅰ型胶原蛋白、NF-κB蛋白及Ⅰ型胶原mRNA、NF-κBmRNA表达与AS发生、发展有着密切关系,该因素的表达增强是致AS的机制之一。Objective：To study the clinical significance and the expression of serum lipid （TC,TG,LDL-C）, type Ⅰ collagen and NF-κB in the occurrence and development of human carotid artery atherosclerosis （CAS）. Method.. The serum lipid （TC,TG,LDL-C） was measured in 18 patients with artery AS confirmed by carotid arteriography, and the plaque was collected by using carotid endarterectomy for pathological studies. The expression of mRNA and protein of type Ⅰ collagen as well as those of NF-κB in plaque were detected with immunohistochemistry, in situ hypbridization technique and CMAIS. Another 6 unexpected dead patients were taken as control. Result： In the human carotid artery atheromatous plaque, the levels of type Ⅰ collagen and NF-κB mRNA were 0. 27±0. 08 and 0.34±0.10, and those of their protein were 0. 49±0. 12 and 0. 58±0.15 respectively, all of which were higher than those in the controls（0.10±0.01, 0.10±0.02, and 0.17±0.02, 0.18±0. 02,0.18±0.02,respectively, P 〈0.05）. Conclusion.. The expression of mRNA and protein of type Ⅰ collagen as well as those of NF-κB might have close relationship with the occurrence and development of AS, and the upregulation of them might be one of the pathogenic mechanisms of AS.
