高效液相色谱法同时测定决明子中大黄素、大黄酚和大黄素甲醚

摘　　要：目的建立同时测定决明子中大黄素、大黄酚和大黄素甲醚的高效液相色谱法。方法采用高效液相色谱法。色谱柱，Diamonsil C18色谱柱（250×4．6mm ID，5μm）；流动相，甲醇-0．1％磷酸溶液（85：15）；检测波长，254nm；柱温，28℃。结果大黄素、大黄酚和大黄素甲醚浓度分别在3．82～38．2μg／mL，7．14～71．4μg／mL和7．81～78．1μg／ml。范围内与峰面积呈良好线性关系（r〉0．9992）；平均回收率（n=6）大黄素为97．56％，RSD=1．21％；大黄酚为98．55％，RSD=0．78％；大黄素甲醚为98．06％，RSD=1．07％。结论该法结果准确，重复性好，可用于决明子中大黄素、大黄酚和大黄素甲醚的同时测定。Objective A HPLC method was developed for simultaneous determination of emodin, chrysophanol and physcion in Semen cassiae . Methods HPLC analysis was performed on a Diamonsil C18 column （250 mm×4.6 mm ID,5μm） with the mixture of methonal and 0. 1% phosphoric acid solution（85 ： 15） as mobile phase. The detection wavelength was set at 254 nm and the column temperature was kept at 28℃ Results The standard curves of emodin, chrysophanol and physcion showed good linearity（ r 〉0. 9992） in the range of 3.82-38.2 μg/ mL, 7.14-71.4μg/mL and 7.81-78.1 μg/mL, respectively. The average recoveries（ n =6） were 97.56%with RSD 1.21 % for emodin, 98.55% with RSD 0.78 % for chrysophanol and 98.06 % with RSD 1.07 % for physcion, respectively. Conclusion The method is accurate and repeatable for determination of emodin, chrysophanol and physcion simultaneously.
