摘 要:目的:观察补肾壮骨中药对绝经后骨质疏松症模型大鼠胫骨细胞水平的客观影响。方法:实验于2003-06/2005-12在解放军广州军区广州总医院实验中心完成。①选用雌性Wistar大鼠40只,9月龄。将大鼠按随机数字表法分为4组:假手术组、模型组、补肾壮骨中药治疗组、降钙素治疗组,每组10只。②麻醉大鼠后,假手术组:切开腹壁,不切除卵巢。其余各组均从背部肋下入路行完整双侧卵巢切除术。补肾壮骨中药治疗组灌胃中药补肾壮骨中药,由广东一方制药厂生产;主要成分:淫羊藿、鹿角胶、龟板胶、骨碎补、巴戟天、淮山药、山茱萸、生地黄、肉桂等;批号:020310,100g/袋;冲剂以温开水溶至浓度为600g/L,用量1mL/(g·d),1次/d。降钙素治疗组:隔次于大鼠双侧前肢外侧皮下注入鲑鱼降钙素注射液(诺华制药厂生产,50IU/支,批号:010208),每次1.04U/kg,每周2次。假手术组、模型组均灌胃等量饮水。③各组均干预3个月,然后取出胫骨上端骨骺部分,做5μm不脱钙切片。用MORPHUMETPYⅡ程序处理数据,计算骨小梁体密度、面密度以及成骨细胞、破骨细胞体密度、数密度。④组间计量结果差异比较采用t检验。结果:实验期间有5只动物因实验操作不当死亡,最终进入结果分析假手术组9只,模型组10只,补肾壮骨中药治疗组8只,降钙素治疗组8只。①骨小梁体密度和面密度:模型组均明显低于假手术组(t=2.215,2.232,P<0.05);补肾壮骨中药治疗组均明显高于模型组(t=2.267~2.308,P<0.05),而与假手术组和降钙素治疗组比较,差异不明显(P>0.05)。②成骨细胞体密度和数密度:模型组均明显低于假手术组(t=2.213,2.215,P<0.05);补肾壮骨中药治疗组均明显高于模型组(t=2.143~2.218,P<0.05),而与假手术组和降钙素治疗组比较,差异不明显(P>0.05)。③破骨细胞体密度和数密度:模型组均明显高于假手术组(t=2.235,2.241,P<0.05);补肾壮骨中�AIM: To explore the objective effect on cellular level of tibia in rats with postmenopausal osteoporosis (PMO) by using Chinese herb of tonifying kidney and strengthening bone. METHODS: The experiment was conducted in the Experimental Center of Guangzhou General Hospital of Chinese PLA between Jnue 2003 and December 2006. ①Forty Wister female rats of 9 months old were selected and randomly divided into 4 groups: sham-operation group, model group, Chinese medicine of tonifying kidney and strengthening bone treatment group (treatment group) and calcitionin treatment group with 10 rats in each group.②Rats in the sham-operation group were cut open the abdominal wall after anesthetization with ovaries remained. Rats in other groups received bilateral oophorectomy via the back and under the fibs. Rats in the treatment group were gastrically perfused of Chinese herb, which was manufactured by Guangdong Yifang Pharmaceutical Corporation and the main components were epimedium, colla comus cervi, colla plastri testudinis, rhizoma drynariae, morinda root, rhiizoma dioscoreae from hena of China, Asiatic cornelian cherry fruit, rehmannia dride rhizome and cassia etc. with the batch number of 020310, 100 g/pack. The powder was made into 600 g/L by dissolving in warm boiled water. The drugs were taken at the dose of 1 mL/g once a day. Rats in the calcitionin treatment group were subcutaneousLy injected with salmon cacitonin see calcimar injection [manufactured by Nuohua Pharmaceurical Company, 50 IU/piece, the batch number was 010208] at the lateral of bilateral anterior limbs for 1.04 U/kg each time and twice per week. Rats in the sham-operatlon group and model group were gastrically perfused of drinking water at the same volume.③All rats were intervened for 3 months. The upper segment of cannon born was obtained and made into 5μm of sections without decalcification.
