脑出血周围组织星形胶质细胞反应与CD34相关性研究

摘  要:目的观察人脑出血周围组织GFAP、CD34的表达,探索脑出血周围组织星形胶质细胞的反应及与微血管的相关性。方法采用HE染色观察星形胶质细胞和微血管的变化,免疫组化技术(两步法)检测胶质纤维酸性蛋白(GFAP)和CD34在星形胶质细胞和微血管的表达。结果脑出血8h其周围组织GFAP表达开始增加(P<0.01),3~5天表达明显增强(P<0.01),7~15天表达达到高峰(P<0.01),以后有所回落,19天时仍高于对照组(P<0.01);CD34阳性反应在脑出血8h开始增加,3~5天其阳性表达数量增多(P<0.01),血管扩张,7~15天达高峰(P<0.01),见大量新生血管,19天时仍比对照组高(P<0.01)。GFAP与CD34消长规律一致(Pearsonr=0.9930,P<0.01,R2=0.9860)。结论脑出血后15天内其周围组织星形胶质细胞增生活跃,并可能促进微血管重建与扩张,两者在组织损伤修复过程中起重要作用。Objective To observe the expression of GFAP and CD34, the reaction of astrocytes and the correlation with microvessels in the surrounding areas after cerebral infarction in human was researched. Methods HE and immunohistochemistry were used to observe the changes of glial fibrillary acidic protein (GFAP) and CD34 in the surrounding areas after cerebral infarction in human. Results The expressions of GFAP and CD34 increased in 8h (P 〈 0.01 )and reached the peak in 7 - 15 days (P 〈 0.01 ) , their expressions had increasing trend within 15 days after cerebral infarction in human, after 15 days the exPressions of GFAP and CD34 decreased and was higher than that in controlled group,the expression between GFAP and CD34 showed positive correlation ( Pearson r = 0. 9930,P 〈 0.01, R^2 = 0. 9860). Conclusions Within 15 days after cerebral infarction astrocytes abnormally activated and possiblly encouraged microvessels to proliferate and dilatation, which suggested that astrocytes and microvessels played an important role in the process of injury and repair.
