摘 要:目的:研究黄花倒水莲总苷(PTS)对由四氯化碳、氨基半乳糖与酒精造成的肝损伤的预防与治疗作用。方法:小鼠分别给予PTS与双联苯酯10d后分别腹腔注射四氯化碳与氨基半乳糖,测定血清中肝功能指标与肝组织中MDA含量。通过大鼠持续酒精灌胃造成酒精性脂肪肝模型,模型建立后给予PTS与非诺贝特,测定血清中相关肝功能指标与血清中和肝组织中脂质含量的变化。对肝组织进行HE染色,光镜下进行组织病理学检查。结果:小鼠给予PTS与联苯双酯10d后腹腔分别注射四氯化碳与氨基半乳糖,与模型组相比,血清GOT与GPT水平显著性降低,肝组织中MDA含量显著性降低。酒精灌胃5周后,PTS与非诺贝特给药3周后,与模型组相比血清中GOT水平显著性下降,血清与肝组织中TC与TG含量均显著性降低,血清中MDA与LDL含量均显著性降低,SOD值显著性升高,肝组织病理组织学变化改善。结论:PTS对由四氯化碳与氨基半乳糖造成的急性肝损伤有治疗作用;对酒精性脂肪肝有保护作用,PTS改善了模型的脂质代谢并降低了由自由基引起的氧化作用。AIM:To discuss the protective effect of PTS on liver injury induced by CCl4, galactosamine and alcohol. METHOD: Mice models of acute liver injury were provided with Frs and bifendate., then ip. CCl4 and galactosamine separately. Serum hepatic function indexes and contents of MDA of liver tissue were measured according to the instruction of test kits. Rats models of alcoholic fatty liver were induced by ig alcohol continuously. After the models were established successfidly, provided with PTS and finofibrate. Serum hepatic function indexes and lipid contents of both serum and liver tissue was measured according to the instruction of test kits. Liver tissue were dyed with HE and histopathological changes in liver were observed under light microscope. RESULT: Mice models of acute liver injury were provided with PTS and Bifendate for 10 days, then ip. CCl4 and galactosamine separately, the level of GOT and GFF in serum and the contents of MDA in liver tissue decreased significantly. Rats model of alcoholic fatty liver were established after ig Alcohol for 5 weeks. After treatment by FI'S and Finofibrate for 3 weeks, the level of GOT in serum, contents of TG and TC in both serum and liver tissue, contents of MDA and LDL in serum decreased significantly. Histopathological changes of liver were improved. CONSLUSlON: PTS has therapeutic effect on alcoholic fatty liver and acute liver injury induced by CCl4 and galactosamine. PTS can improve fat metabolism in models and depress the oxygenation of free radicle.
