摘  要:目的:探讨5-脂氧化酶(5-LO)在系统性红斑狼疮(SLE)患者外周血多形核细胞(PMNS)中的表达水平,及其在SLE发病机制中的作用及意义。方法:应用反转录聚合酶链反应(RT-PCR)检测了40例活动期SLE患者和20名正常人PMNS中5-LO的表达水平。结果:SLE患者5-LO的阳性表达率与正常对照组相比差异无统计学意义;但活动期SLE患者PMNS中的5-LOmRNA的平均表达水平明显高于非活动期SLE和正常对照组,而非活动期SLE患者与正常对照组间5-LO的平均表达强度无显著差异。结论:活动期SLE患者中5-LO参与了SLE的发病过程,为特异性5-LO抑制剂或CysLTs受体拮抗剂应用于SLE的治疗提供了理论依据。Objective: To investigate the role and significance of 5-lipoxygenase (5-LO) in the pathogenesis of systemic lupus erythematosus (SLE). Methods: Applying reverse transcription-polymerase chain reaction (RT-PCR) Technique to semiquantitatively analyze 5-LO mRNA expression in the peripheral Polymorphomuclear leukocytes (PMNs) from 40 SLE patients and 20 healthy subjects. Results: Compared with normal controls, there are no statistical difference in the positive rates of 5-LO in SLE. The mean levels of 5-LO mRNA expression in active SLE group were significantly higher than those in non-active SLE group and normal controls respectively. However, there are no significant difference in the mean level of 5-LO mRNA expression between patients with non -active SLE group and normal controls. Conclusion: The levels of 5-LO mRNA expression in active SLE group increase karkedly, which may be involved in the pathogenesis of SLE and provide a valuable scientific basis for the clinical use of 5-LO inhibitors and cysteinyl leukotrienes receptor antagonist.
