健脑益智汤对阿尔茨海默病大鼠学习记忆和氧化应激的影响

摘　　要：目的观察健脑益智汤对AD大鼠学习记忆和氧化应激的影响。方法选出正常老年大鼠48只,随机分为对照组、模型组、安理申组和健脑益智汤大、中、小剂量组。除对照组左侧侧脑室注射5μl生理盐水外,其余各组给予等量聚集态的Aβ25-35建立AD模型,并且各组按规定剂量灌胃3周。采用Morris水迷宫测试各组大鼠学习记忆能力,并检测血清和脑组织MDA含量和SOD活力。结果健脑益智汤大、中剂量组较安理申组能更有效地减少AD大鼠MDA含量(P<0.05)、增加SOD活力;而对AD大鼠学习记忆能力的改善作用二者无明显差别。结论健脑益智汤可以通过影响自由基水平对阿尔茨海默病大鼠有一定的防治作用。Objective To investigate protective effects of JiannaoYizhi decoction on the model rats with Alzheimer＇s disease. Methods To selecte male normal old rats that was randomly divided into control group, model group,Aricept group and big,moderate, small dose of JiannaoYizhi decoction groups. Except the control group was injected 5 μl sodium chloride into the left lateral ventricle of old rats,other groups was injected the same dose aggregated Aβ25 - 35 to make AD models. Morris test was used to determine the ability of learning and memory. The activity of superoxide dismutase and the content of malondialde hyde in brain and serum were determined. Results High and middle doses of JiannaoYizhi decoction were able to improve the activity of superoxide dismutase and decrease the content of malondialde hyde（P〈 0.05） compared with Aricept, but there was no significant difference between JiannaoYizhi decoction and Aricept for improving the decreased ability of learning and memory induced by β-amyloid25 - 35. Conclusion JiannaoYizhi decoction may have preventive and therapeutic effects through influence on the free radical levels in rats with AD.
