新生大鼠骨源性间充质干细胞向多巴胺能神经元样细胞的分化

摘　　要：目的探讨体外培养的骨源性间充质干细胞向神经元样细胞的分化。方法采用体外骨植块法培养骨源性的间充质干细胞(mesenchymal stem cells,MSCs),传代。用含0.5mmol/L IBMX,GDNF(10 ng/ml)和IL-1β(100 pg/ml),中脑神经胶质细胞条件培养基和中脑神经膜碎片诱导其向多巴胺能神经元样细胞分化,免疫细胞化学检测神经细胞的表面标志NSE、MAP-2和TH。结果原代骨源性间充质干细胞含有多种细胞形态,传代后细胞成均一的梭形。诱导前,对照组少量细胞表达NSE,不表达MAP-2和TH。诱导后间充质干细胞分化成表达NSE、MAP-2和TH的细胞。结论骨源性间充质干细胞分化成神经元样细胞,表明其可跨胚层分化,同时为细胞移植治疗中枢神经系统疾病提供一种新的细胞来源。Objective To explore to differentiation of mesenehymal stem eells（MSCs） derived from bone into neuron - like cells.Methods MSCs derived from bone were cultured and passaged using bone explants. Then MSCs were induced to differentiate into neuron - like cells by 0.5mmol/L IBMX, GDNF（10ng/ml）, IL-1β（100pg/ml）, mesencephalic glial-cell-conditioned medium and flash- frozen mesencephalic membrane fragments. The markers of neuron, NSE.MAP- 2 and TH were detected using the methods of immunocytoehemistry. Results Before being induced, few of MSCs expressed NSE and no MSCs expressed MAP-2and TH in control group. After being induced, MSCs derived form bone differentiated into neuron-like cells which expressed NSE.MAP-2 and TH. Conclusion It showed that MSCs derived from bone may differentate into cells of another embryonic layer and supply a new cell source for cells transplantation to treat central nervous system disease that MSC derived form bone differentiated into neuron-like cells.
