摘 要:以盐敏感型山定子实生苗和耐盐型珠眉海棠组培苗为材料,采用灌注离心技术研究了叶片质外体和共质体中Na+和Ca2+浓度的变化.结果表明:随盐胁迫强度的加强,叶片水势下降;叶片Na+含量、质外体和共质体中Na+浓度升高,珠眉海棠明显低于山定子;叶片Ca2+含量、共质体Ca2+浓度随盐胁迫的增加而升高,但珠眉海棠高于山定子,50 mmol/L NaCl胁迫对质外体Ca2+没有明显影响,100 mmol/L NaCl胁迫下增加,珠眉海棠低于山定子;叶片共质体与质外体中Na+浓度的比值,珠眉海棠明显高于山定子,说明在盐胁迫下珠眉海棠具有较强的离子区域化能力,离子区域化是珠眉海棠的主要耐盐机制.Salt-sensitive Malus baccata seedlings and salt-tolerant in vitro Malus zumi seedlings were used to study Na^+ concentrations in their leaf apoplast and symplast by vacuum infiltration and centrifugation. It was shown that as salt stress became intensified,the leaf water potential declined;the Na^+ contents in the leaves and the Na^+ concentrations in the apoplast and symplast increased and M. zumi showed a high increment in the Na^+ contents and concentrations than M. baccata ;as salt stress became intensified,the Ca^2+ contents in the leaves and the Ca^2+ concentrations in the apoplast and symplast rose and M. zumi showed a high increment in the Ca^2+ contents and concentrations than M. baccata ;50 mmol/L NaCl stress did not show remarkable influence on the Ca^2+ concentrations in the apoplast and 100 mmol/L NaCl stress increased the concentrations, more in M. zumi than in M. baccata ; the ratio between the Na^+ concentrations in the leaf symplast and apoplast was higher in M. zumi than in M. baccata ,indicating that M. zumi had a higher compartmentalizing capacity and compartmentalization was its main salt-tolerance mechanism.
