摘 要:目的:观察比较聚乙烯、一次性使用静脉留置针、血浆袋、输血管4种试验材料与人血清接触后,对人补体C3含量的影响。方法:实验于2004-04/2005-02在上海生物材料研究测试中心血液相容性检测室完成。①取健康人新鲜静脉血,获得正常血清。②将0.6cm2聚乙烯、一次性使用静脉留置针、血浆袋、输血管4种试验材料加入0.1mL血清,(37±1)℃水浴中孵育60min。空白血清作为阴性对照材料,橡胶手套作为阳性对照材料。③通过免疫比浊法来测定补体C3的含量的变化。结果:每组材料重复3管。①聚乙烯、一次性使用静脉留置针、血浆袋、输血管4组实验材料接触血清后的补体C3含量均略有减少[依次为(1.049±0.001),(1.066±0.012),(1.053±0.022),(1.100±0.090)g/L]。②4组材料C3浓度值与阴性对照组[(1.075±0.020)g/L]相比差异无显著性(P>0.05)。③阳性对照组则显示材料接触血清后的补体C3含量[(0.350±0.058)g/L]显著低于4组材料及阴性对照组。结论:①聚乙烯、一次性使用静脉留置针、血浆袋、输血管4组实验材料对机体的补体系统均无明显的不良影响。②补体C3含量测定的方法简便,结果明确,可推荐作为生物材料的免疫学评价方法之一。AIM: To study the effects on contents of human complement C3 by contacting four kinds of test materials: polythene, once venous detain, blood plasma sack and race track with human serum. METHODS: The experiment was conducted in the Testing Department of Blood Compatibility, Shanghai Biomaterials Research & Test Center from April 2004 to February 2005. ① Fresh venous blood was drawn from healthy people to obtain normal serum.②0.6 cm2 of polythene, once venous retention needles, blood plasma sack and race track were contacted with 0.1 mL serum to incubate in water for 60 minutes at (37±1)℃. The blank serum were taken as negative control, and a latex glove was taken as the positive control. The immunoturbidimetry was adopted to determine the changes in contents of C3. RESULTS: Materials of each group was repeated for 3 tubes. ①The contents of C3 slightly decreased after contacting with polythene, once venous detention needles, blood plasma sack and race track [which were respectively ( 1.049±0.001 ), ( 1.066±0.012 ),(1.053±0.022), ( 1.100±0.090)g/L]. ②There were no significant differences in concentration of C3 between four material groups and negative control group [(1.075±0.020) g/L], (P〉0.05). ③ The contents of C3 in complement of positive control group [ (0.350±0.058) g/L] after contacting with serum was obviously reduced than that of four material groups and negative control group. CONCLUSION: ①There are little harmful effects of polythene, once venous detention needles, blood plasma sack and race track on human complement system. ②The measurement of complement C3 is convenient with definite results, which can be recommended as one of the immunological evaluations for biomaterials.
