摘 要:目的:探讨甘草抗病毒有效部位(GC3-1-4)体外抑制呼吸道合胞病毒(RSV)的作用。方法:采用煎煮法、乙醇沉淀法和柱层析法分离提取GC3-1-4。以病毒唑为阳性对照药物,采用细胞培养技术观察GC3-1-4抑制RSV作用。结果:GC3-1-4的TC50为0.45mg/ml,抑制RSV的EC50为41.32μg/ml,TI为10.89。GC3-1-4对RSV的抑制作用存在量效关系;感染RSV后0h、2h、4h、6h、8h加药,GC3-1-4在RSV的活性有抑制作用。化学实验证实GC3-1-4不含甘草酸或多糖类物质。结论:GC3-1-4在Hela细胞中对RSV有明显的抑制作用。Objective:To study the effect of anti-respiratory syncytial virus(RSV)on an active compound(GC3-1-4)from glycyrrhiza in vitro.Methods:The GC3-1-4 was isolated from glycyrrhiza,by means of hot water extraction,ethanol precipitation and column chromatography.The activity of GC3-1-4 against RSV was observed by cell culture while Ribavirin was used as a positive control.Results:The TC_(50) of GC3-1-4 was 0.45mg/mL,the EC_(50) of GC3-1-4 against RSV was 41.32μg/mL and TI was 10.89 The GC3-1-4 achieved a concentration-dependent inhibition of the replication of RSV in vitro.The GC3-1-4 showed and active effect against RSV replications in vitro when the GC3-1-4 was added at Oh,2h,4h,6h,8h,after virus infection.Chemical tests showed that GC3-1-4 was not glycyrrhizic acid or polysaccharide.Conclusion:The study suggested that GC3-1-4 was an obvious inhibitor to RSV in Hela cell culture.
