舒痛安汤对吗啡依赖大鼠脑β-EP及血清性激素的影响

摘  要:目的:观察由黄芪、党参、钩藤等组成的舒痛安汤(STA)对吗啡依赖大鼠脑β-EP及血清性激素的影响。方法:采用大鼠戒断模型,观察舒痛安汤对吗啡依赖大鼠下丘脑和血浆β-内啡肽(β-EP)、结状神经节和孤束核P物质(SP)以及血清性激素的变化。结果:(1)吗啡依赖大鼠下丘脑β-EP的含量均较正常大鼠降低,舒痛安汤高剂量组可增高下丘脑β-EP的含量(P<0.01),STA中、高剂量组可增高血浆β-EP的含量(P<0.01,P<0.05);吗啡依赖大鼠结状神经节和孤束核SP的含量升高,STA高剂量组可降低结状神经节和孤束核SP的含量(P<0.01,P<0.05)。(2)吗啡依赖雌性大鼠血清卵泡刺激素(FSH)、雌二醇(E2)、泌乳素(PRL)均低于正常对照组,STA中、高剂量组均可增高血清FSH、E2、PRL的含量(P<0.01,P<0.05);(3)吗啡依赖雄性大鼠睾丸酮(T)含量显著降低(P<0.01),STA中、高剂量组均可增高雄性大鼠血清T的含量(P<0.01),而对大鼠血清黄体生成素(LH)、PRL的含量无显著影响。结论:STA可能通过改善吗啡依赖大鼠下丘脑-垂体-肾上腺的变化,消除其戒断症状,从而达到治疗的目的。Objective: To observe the effect of Brain and serum sex hormone secreted - EP in morphine dependence rats with Shu TongAn Decoction (STA). Method:To detect the change of β -endorphin in hypothalamus and plasm, substance P in ganglion nodosum and nucleus tractus solitarii and incretion hormones of Serum sex hormone in morphine dependence rats. Result: ( 1 )The level of - endorphin hypothalamus of addicted mouse was reduced obviously than that in normal rats (P 〈 0.01 ). The high dose group of Jiedutuoying Decoction can improve the level of β - endorphin of morphine dependence rats( P 〈 0.01 ,P 〈 0.05 ). The level of β - endorphin in plasm can be improved in the moderate and high dose groups of STA( P 〈 0.01 ,P 〈 0.05 ) . The level of substance P in ganglion nodosum and nucleus tractus solitarii in addicted mouse was increase obviously than that in normal rats and STA can moderate the level in high dose group (P 〈 0.01, P 〈 0.0.5 ). (2) The level of FSH, E2 and PRL in addicted rats was reduced obviously than that in normal rats. The level of FSH, E2 and PRL in plasm can be improved in the moderate and high dose groups of STA(P 〈 0.01 ,P 〈 0.05 ). ( 3 ) The level of testosterone in addicted rats was reduced obviously than that in normal rats and STA can improved the content in the moderate and high dose groups (P 〈 0.01,P 〈 0.05 ), but no effect on LHand PRL. Conclusion:The medicine maybe improve the axis of hypothalamus - pituitary - adrenal gland and eliminate the addicted symptom of rats with morphine.
