摘 要:目的探讨生长受限胎鼠脑组织神经细胞凋亡率变化,以及孕期应用钙离子拮抗剂尼莫地平干预对神经系统损伤是否具有保护作用。方法SD孕鼠随机分为3组:①组Ⅰ:行假手术;②组Ⅱ,采用缩窄子宫动脉法建立FGR模型;③组Ⅲ:建模方法同组Ⅱ,术后予尼莫地平治疗。电子天平对新生仔鼠脑组织称重,比较各组仔鼠脑重。脑组织切片,HE染色,观察脑组织形态学变化。通过TUNEL法检测各组新生仔鼠皮层和海马CA1区神经细胞凋亡。结果①组Ⅰ仔鼠脑重为0.227±0.168g,明显高于组Ⅱ(0.208±0.192g,P<0.01)以及组Ⅲ(0.215±0.139g,P<0.01),而Ⅱ、Ⅲ两组之间差异无统计学意义(P>0.05)。组Ⅱ部分仔鼠脑组织皮层变薄,存在出血、水肿灶,而组Ⅰ和组Ⅲ仔鼠未见明显变化。②组Ⅲ仔鼠皮层神经细胞凋亡率显著低于组Ⅱ(分别为2.98±0.54个/HP和5.05±1.17个/HP,P<0.01),但高于组Ⅰ(2.3±0.69个/HP,P<0.01)。③组Ⅲ仔鼠海马CA1区神经元凋亡率中位数为0.83个/HP,低于组Ⅱ(1.50个/HP,P<0.01),高于组Ⅰ(0.50个/HP,P<0.05)。结论钙离子拮抗剂尼莫地平孕期治疗对生长受限仔鼠神经系统损伤有保护作用。Objective To study the properties of cerebral histomorphology and neuron apoptosis in growth restricted rat fetus, and whether nimodipine treatment during gestation can attenuate nervous system injuries. Methods Pregnant SD rats were randomly divided into 3 groups. Sham operation was exerted for rats in group Ⅰ, while bilateral uterine arteries were partially ligated in group Ⅱand Ⅲ. Nimodipine (0. 2mg/kg/d) was administrated subcutaneously for the group Ⅲ. Each fetus whole brain was weighted by electronic balance. Cerebral morphological changes were examined by light microscopy. Neuron apoptosis in cortex and hippoeampus was detected by TUNEL. Results ①The brain weight of group Ⅰ (0. 227±0.. 168g) was significantly higher than group Ⅱ (0. 208±0. 192g,P〈0. 01) and m (0. 215±0. 139g,P〈0. 01), but there was no diffirence between these two group.②The ratio of neuron apoptosis in cortex of group m was significantly lower than that of groupⅡ (2. 98±0. 54/HP vs 5. 05±1. 17/HP, P〈0. 01) ,but higher than that of group Ⅰ (2. 3±0. 69/HP, P〈0. 05). ③ The median ratio of neuron apoptosis in hippoeampus CA1 in group Ⅲ was 0. 83/HP, significantly lower than group Ⅱ (1. 50/HP, P〈0. 01), but higher than group Ⅰ (0. 50/HP P〈0. 01). Conclusion Calcium channel antagonist treatment during gestation has a protective effect on neural injury induced by FGR.
