急性出血坏死性胰腺炎小鼠辅助性T细胞亚群1/2的变化规律及中药干预作用研究

摘　　要：目的:探讨急性出血坏死性胰腺炎小鼠早期出现辅助性T细胞(T h)亚群T h1/T h2变化规律,观察参麦注射液及黄芪注射液的干预作用。方法:65只昆明小鼠随机分为假手术组5只,胰腺炎组、参麦治疗组、黄芪治疗组各20只。采用腹腔连续注射雨蛙素50μg/kg,每次间隔1 h,连续注射6次后,向腹腔内注射脂多糖造成小鼠急性出血坏死性胰腺炎模型。治疗组分别在制模后向腹腔内注射参麦注射液或黄芪注射液,于末次注射完毕后2、6、12和24 h,分别测定各组动物血浆淀粉酶及脾细胞的T h1/T h2比值,观察胰腺及小肠病理变化。结果:胰腺炎组制模后6 h胰腺病理及血浆淀粉酶提示出现急性出血坏死性胰腺炎,同时出现脾细胞T h1/T h2比值失衡(P<0.01),并逐渐加重,在24 h时甚至出现倒置。而中药治疗组此种变化较轻,在制模后24 h参麦注射液较黄芪注射液疗效更显著(P<0.05)。结论:小鼠急性出血坏死性胰腺炎早期出现免疫失衡,参麦注射液和黄芪注射液对此有调节作用。Objective： To investigate regularity of changes of helper T cell （Th）subgroup Th1/Th2 in mice with acute hemorrhage necrotizing pancreatitis （AHNP）, and observe the interference effect of Shenmai injection （参麦注射液） and Huangqi injection （黄芪注射液）. Methods ： Sixty-five Kunming mice were randomly divided into sham operation group （n ： 5）, pancreatitis model group （n = 20）, Shenmai injection group （n = 20） and Huangqi injection group （n = 20）. The AHNP model was established by intraperitoneally injecting caerulein 50μg/kg every 1 hour for 6 times and then intraperitoneal injecting lipopolysaccharide 10 mg/kg at the last time. The two treatment groups received Shenmai injection or Huangqi injection intraperitoneal injection respectively. The ratio of spleen cell Th1/Th2 and plasm amylase were detected at 2, 6, 12 and 24 hours after the last injection respectively with the sham operation group included, and the pathological changes of pancreatic gland and small intestine were observed by microscope. Results： The AHNP model was confirmed 6 hours after the last injection by pathological changes and plasm amylase, and the ratio of spleen cell Th1/Th2 was decreased gradually （P〈0. 01）, and inversed after 24 hours, these changes were milder in the two treatment groups （all P〈0.01）. Conclusion. An immune unbalance occurs in the early stage of AHNP, Shenmai injection and Huangqi injection can modulate it.
