摘 要:考察外源性的1,6-二磷酸果糖(FDP)对心肌细胞内游离Ca2+浓度和pH值的影响。用荧光探针(Fura-2,BCECF)技术同时测定成年大鼠心肌细胞内游离Ca2+浓度和pH值,并观察其在缺氧/复氧条件下的变化。结果:在缺氧/复氧过程中,胞质游离Ca2+浓度缓慢升高,从基础值130±29.5nmol/L1升至742±154nmol/L,胞质pH值则从7.12±0.08降至6.71±0.13。在含有5mmol/LFDP的细胞悬液中,则胞质游离Ca2+上升幅度明显低于对照组(P<0.05),在复氧结束时的最高值为538±193nmol/L,但胞质pH值的变化与对照组比较无显著差别。结论:FDP可以减轻由缺氧/复氧所造成的心肌细胞内游离Ca2+浓度增高的程度,而对胞质pH值的变化无明显影响。PURPOSE To study if exogenous fructose-1,6-diphosphate (FDP)exerts homeostatic regulatory effect on intracellular Ca2+ ([Ca2+]i) elevation and pH ([pH]i) changes during hypoxia/reoxygenation in isolated rat myocytes.METHODS Utilizing fluorescent Ca2+and H+ indicators (Fura-2 and BCECF)techniques to study the effect of FDP on disturbances of [Ca2+]i and [pH]i in isolated rat cardiocytes simultaneously which were exposed to hypoxia succeeded by reoxygenation.RESULTS Exposure to hypoxia followed by re-oxygen produced a sustained increases in [Ca2+]i from 130±29.5 nmol/L upto 742±154 nmol/L,and a slow decrease in [pH]i from 7.12± 0.08 down to 6.71± 0.13.FDP (5 mmol/L) significantly attenuated the characteristic damage reflected by stepping-up increase in [Ca2+]i (538±193 nmol/ L,P<0.05), but did not prevent the occurrence of intracellular acidosis (6.74±0.18, P>0.05). CONCLUSIONS FDP prevents the increase in [Ca2+]i level in myocytes isolated from rat heart during hypoxia/reoxygenation, but unaccompanied by correction of acidosis.
