转基因741杨叶片对卡那霉素适应性分析

摘  要:采用不同浓度卡那霉素溶液处理转基因741杨株系Pb17、Pb29及未转基因741杨一年生枝条,测定它们在不同处理下的各项生理指标,探索作为选择标记基因的新霉素磷酸转移酶基因(nptⅡ)在转基因植株中的表达情况。结果表明,随着卡那霉素处理浓度的提高,各转基因株系及对照的酶促防御系统SOD、POD活性降低,净光合速率、荧光动力学参数下降,但Pb29、Pb17下降幅度均低于对照,证明nptⅡ基因在转基因741杨叶片中与目的基因同步表达,使转基因株系具备抗卡那霉素的能力。One-year-old twigs of transgenic poplar 741 clones (Pb17, Pb29) and non-transgenic poplar 741 (control) were treated with differem concentrations of kanamycin to determine every physiological index for each treatment and explore the expression of neomycin phosphotransferase gene (npt Ⅱ) in the transgenic plants as a selective marker. Result showed that with the concentration of kanarnycin increasing, SOD and POD activities of enzyme recovery system reduce, and net photosynthetic rate and kinetic parameters of fluorescence induction decrease for the transgenic clones and the control, while the reduction of Pb17 and Pb29 is lower than that of the control. It indicates the synchronous expression of npt Ⅱ gene and the target gene, which results in the resistance ability of the transgenic plants to kanamychin.
