摘 要:通过引物设计、PCR、基因克隆、测序,获得了10种鸢尾属植物和1个外类群种的ITS序列。鸢尾属植物ITS1区长度219~243bp,5.8S序列长度均为165bp,ITS2区长度变异范围为196~239bp。5.8S序列过于保守,保守位点占81.9%。ITS序列对于区分鸢尾属植物组级的较大分类等级较好。用近缘属射干属的Belamcanda chinensiss作为外类群,用邻位相接法(NJ)和最大简约法(MP)进行聚类分析,得到了较一致的系统树,且分析表明,ITS序列对于建立属间的分类等级更加有效。结合Genebank中的中国鸢尾属植物的序列,比较支持赵毓棠建立的中国鸢尾属植物组以上的分类系统。ITS sequences of ten kinds of Iris plants and an outgroup were obtained by primer design, PCR, gene cloning, sequencing and cluster analysis. The ITS1 section length is 219~243 bp, 5.8 S sequence length is 165 bp in the species of Iris, and the ITS2 section length varies from 196 bp to 239 bp. The 5.8 S sequence is too conservative, and the conservative sites occupy 81.9%. The ITS sequence has many advantages in the discrimination of the large classification grade such as Iris subgenus-joining. A more consistent systematic tree was obtained by cluster analysis with maximum parsimony method and neighbor method taking relative Belamcanda chinensis as an outgroup. The analysis indicates that the ITS sequence is more effective in establishing classification rank among genera. Considering the sequence of Chinese Iris species in Genebank, the result supports the taxonomy system of the upper group of Chinese Iris species, established by Zhao Yutang.
