摘  要:目的:建立大鼠肾脏缺血再灌注损伤(IRI)的缺血后处理模型。方法:行右肾切除,左肾蒂分别钳夹0分钟、30分钟、45分钟、60分钟,再灌注24小时后,根据肾功能检测指标确定大鼠IRI模型;大鼠随机分为4组,对照组、缺血再灌注组、缺血后处理1组和缺血后处理2组,再灌注24小时后根据肾功能检测指标和肾组织病理损伤程度分级的变化确定缺血后处理模型。结果:右肾切除,左肾蒂钳夹60分钟,再灌注24小时后,肾脏功能检测指标显著升高(P<0.05),确定IRI模型肾蒂钳夹时间为60分钟;与I/R组相比,经缺血后处理-1干预后,大鼠肾脏功能检测指标显著降低(P<0.05),肾组织损伤明显改善,病理损伤分级明显降低(P<0.05);经缺血后处理-2干预后,大鼠肾脏功能和组织损伤程度无明显改善(P>0.05)。结论:采用缺血后处理-1的方法可以成功建立大鼠肾脏IRI的缺血后处理模型。Objective: To construct the model of ischemic postconditioning (Pos - con) on renal ischemia - reperfusion injury (IRI) in rat. Methods: After right nephrectomy, the renal pedicles of rats were clamped 0min (control), 30min, 45min and 60min with micro- artery clamps, respectively.After 24h of reperfusion, the blood urea nitrogen (BUN) and sertum creafine (Scr) were measured to decide the IRI model.40 Wistar rats were randomized into 4 groups: group A (Control), group B (I/ R), group C (Pos - con1) and group D (Pos - con2) .After 24h reperfusion without control, the blood samples were taken through the vena cava and the renal samples were taken too. Serum urea nitrogen and creafine were measured. The renal samples were examined by HE stain under light microscopy. Results: Compared with the control group, 60rain ischemia and 24h reperfusion can increased the BUN and Scr significantly (P 〈 0.05) .Compared with IRI, the BUN and Scr of Pos- Conl were decreased remarkably ( P 〈 0.05), the injury of kidney was attenuated and the pathological scale was reduced significantly ( P 〈 0.05) .However, Pos- con2 didn' t take the same progression. Conclusion: Stable model of ischemic postconditioning on renal IRI in rat can be prepared by Pos- conl.
