摘  要:目的观察模拟空间发射环境高+Gx对猴口腔黏膜上皮组织结构和热休克蛋白质70(HSP70)表达的影响。方法以雄性猕猴为对象,随机分为4组,对照组承受+1Gx/300s的超重作用;实验组根据承受过载峰值的大小又分为3组,分别为+15Gx/200s、+18Gx/165s和+21Gx/140s。采用常规组织病理学、免疫组织化学方法,观察猴口腔黏膜上皮细胞组织结构和HSP70表达。结果组织病理学观察显示,对照组和各实验组均未见明显改变;免疫组织化学观察显示,对照组口腔黏膜上皮部分基底细胞和少量棘细胞呈浅棕色,为HSP70弱阳性表达,实验组口腔黏膜上皮基底细胞和棘细胞呈深棕色,为HSP70强阳性表达,高+Gx实验组之间口腔黏膜上皮HSP70表达无明显差异。结论高+Gx环境中猴口腔黏膜上皮组织结构未见改变,上皮细胞HSP70表达明显增强。Objective To observe the structural changes of oral mucosal epithelial tissue of Rhesus macaque following high sustained +Gx loads. Methods Nine male Rhesus macaques were randomly divided into four groups. Control group underwent +1 Gx/300 s on animal centrifuge, and experimental groups 1, 2 and 3 underwent +15 G J200 s, +18 Gx/165 s and +21 Gx/140 s respectively. Oral mucosal tissue was fixed with 10% buffered formaldehyde and embedded with paraffin, and histopathological changes were observed with microscope. The expression of HSP-70 in oral mucosal tissues of the animals was detected by immunohistochemical PicTure^TM method. Results Gross inspection and histological observation showed that no significant pathological changes were found in the oral mucosal tissues in all 3 experimental groups of the animal. Immunohistochemical staining demonstrated that expression of HSP-70 in the basal and spinous cells of oral mucosal epithelia in 3 experimental groups were more intensive than that in control group. The differences of HSP-70 expression among 3 experimental groups were non-significant. Conclusions High sustained high Gx load will not lead to significant pathological changes in oral mucosal epithelia, but can induce enhanced expression of HSP-70 in the tissue.
