摘 要:目的建立毛细管等速电泳(CITP)分析血清脂蛋白的方法。方法用盐酸和丙氨酸组成的不连续缓冲体系分离荧光标记的脂蛋白颗粒;各亚组分的相对峰面积(RPA)结合血清总胆固醇反映各脂蛋白胆固醇的含量。结果本法可在6min内将血清脂蛋白分出9条带,其中1—4为HDL,5为CM,6为VLDL/LDL,7—9为LDL。各组分的RPA批内CV为3.17%~13.55%,批间CV为4.55%~15.33%;血红蛋白〈20g/L、胆红素〈342μmol/L、清蛋白〈70g/L对结果无影响;HDL-C、LDL-C与均相法相关(r分别为0.87和0.84,P〈0.001)。结论CITP可快速、灵敏分离定量血清脂蛋白及其胆固醇含量,可用于临床分析脂质代谢异常。Objective To develop a method for separation of serum lipoproteins by capillary isotachophoresis(CITP). Methods Fluorescence-tagged particles of lipoproteins were separated by discontinuous buffer system contained hydrochloric acid and alanine. HDL cholesterol and LDL cholesterol were quantified according to the percentage of relative peak area(RPA) combined with the total cholesterol in serum. Results Serum lipoproteins were separated into 9 subfractions in 6 minites. Within-run CV of RPA displayed as 3.17% to 13. 55% and between-run CV as 4.55% to 15.33%. There was no significant interference for the results when the concentrations of hemoglobin,bilirubin and albumin in sera were lower than 20 g/L,342 μmol/L and 70 g/L respectively. The results of HDL cholesterol and LDL cholesterol detected by CITP correlated with those detected by homogeneous phase assay. Conclusions CITP is a new method for separation and quantification of serum lipoproteins and cholesterols, and the results may rapidly, sensitively reflect status of lipid metabolism.
