摘 要:目的通过观察体外冲击波干预骨髓间充质干细胞(hMSCs)后细胞形态学的变化,探讨体外冲击波(ESW)促进成骨的机理。方法用5KV、100频次的理想体外冲击波干预第1代骨髓间充质干细胞,分别应用倒置相差显微镜投射电镜及HE染色观察细胞形态结构变化。结果体外冲击波对hMSCs增殖的影响显著,第3—5代hMSCs核分裂相增多,核浆比加大,增殖高峰提前,细胞簇集融合,呈放射状扩展;第7—9代细胞体积增大明显,内质网扩张成池,高尔基器发达,条带状交错成片,基质堆积成束,方向性明显,具有成熟细胞的表现;第11代仍有呈对称分裂细胞,表现出定向成骨分化的潜能。结论ESW干预能够提高hMSCs的增殖和分化能力。Objective To explore the morphological changes of human mesenchymal stem cells (hMSCs) after exposure to extracorporeal shock wave (ESW). Methods An optimal experimental energy dose of ESW was applied to hMSCs of passage one. The morphological changes were examined by using inverted phase contrast microscope ( IPCM), transmission electron microscope and cytochemical staining. Results ESW of 5 kV and 100 times could raise the viability and proliferation of the cells. In the hMSCs of passage 3 - 5, the multiplicatire division was speeded up, and the cells in the nuclear fission phase were increased. In the hMSCs of,passage 7 - 9, the volume of the cells increased, and the endoplasmic reticulum was expanded. The Golgi's apparatus was developed and the matrix of the cells was bunchy. In the hMSCs of passage 11, the multiplicative division was also detected. Conclusion The proliferation and differentiation of the hMSCs can be improved by the treatment of ESW.
